Repression of the PRELP gene is relieved by histone deacetylase inhibitors through acetylation of histone H2B lysine 5 in bladder cancer

Abstract: Background Proline/arginine-rich end leucine-rich repeat protein (PRELP) is a member of the small leucine-rich proteoglycan family of extracellular matrix proteins, which is markedly suppressed in the majority of early-stage epithelial cancers and plays a role in regulating the epithelial–mesenchymal transition by altering cell–cell adhesion. Although PRELP is an important factor in the development and progression of bladder cancer, the mechanism of PRELP gene repression remains unclear. … Show more

“…The protein functions as a molecule anchoring basement membranes to the underlying connective tissue. This protein has been investigated in the pathogenesis of Hutchinson-Gilford progeria, psoriatic and rheumatoid arthritis, bladder cancer and respiratory tract infections [50]. ADORA2B gene encodes an adenosine receptor that is a member of the G protein-coupled receptor superfamily.…”

“…Among chromatin remodelers, histone deacetylases (HDACs) can create a condensed chromatin structure that significantly mediates transcriptional repression by removing acetyl groups from histones, which reduces their electrostatic charges, thus enhancing DNA binding . Extensive research studies have indicated a close association between aberrant levels of HDACs and the development and progression of various diseases, including cancer. Therefore, accurate monitoring and measurement of HDACs are crucial for the diagnosis, treatment, and screening of HDAC-related cancer …”
