Test No. 475: Mammalian Bone Marrow Chromosomal Aberration Test

“…If a preliminary range-finding study is performed because there are no suitable data already available to aid in dose selection, it should be performed in the same laboratory, using the same species, strain, sex, and treatment regimen to be used in the main study (10). The study should aim to identify the maximum tolerated dose (MTD), defined as the highest dose that will be tolerated without evidence of study-limiting toxicity, relative to the duration of the study period (for example, by inducing body weight depression or hematopoietic system cytotoxicity), but not death or evidence of pain, suffering or distress necessitating humane euthanasia (11).…”

“…Si l'on procède à une étude préliminaire de détermination des doses à administrer parce qu'on ne dispose pas de données fiables pour orienter le choix des doses, cette étude préliminaire doit être effectuée dans le même laboratoire, en utilisant une espèce, une souche, un sexe et un régime de traitement identiques à ceux de l'étude principale, conformément aux recommandations relatives aux études de détermination des doses (12). L'étude devra avoir pour objectif de déterminer la dose maximale tolérée (DMT), définie comme la dose entraînant de légers effets toxiques dans le cadre de la durée de l'étude (par exemple, en induisant un comportement ou des réactions anormales, une baisse mineure du poids corporel ou une cytotoxicité du système hématopoïétique) mais ne provoquant pas la mort ou des signes de douleur, de souffrance ou de détresse imposant une euthanasie de l'animal (13).…”
