Replication of Rhinovirus RNA

Abstract: SUMMARYWe have studied the synthesis of virus RNA in human embryo lung cells infected with rhinovirus type 2. The three species of RNA in extracts of infected ceils are, in order of decreasing electrophoretic mobility, single-stranded RNA, replicative form and replicative intermediate. The kinetics of synthesis of these RNA species were investigated. The electrophoresis of mixtures of single-stranded RNA synthesized early and late in infection showed no differences in size. This observation was confirmed with … Show more

“…Intact virion t{NA is infectious (39, 57, 87, 11.4), and sediments somewhat slower than the RNA of many other picornaviruses (9,89). In contrast to the slower sedimentation rate, it has been reported that HRV-2 I~,NA migrates behind poliovirus RNA in gel electrophoresis (57). Thus, depending upon the method used, the molecular weight of rhinovirus type2 ]gNA has been estimated between 2.2 and 2.8 × 106.…”

“…Replication of HRV-2 or HRV-14 is complete in 10--14 hours at 33--35 ° C in monolayer or suspension cultures of human heteroploid cells or monolayers of human diploid cells (34,39,57,126,135). HRV-20 required 15 hours for maximum yields in WI-38 cells at 36 ° C (112).…”

“…29.4 ) [ 129 ] with membrane-associated replication structures containing double-stranded RNA (dsRNA) replicative intermediates (RI) which are formed in cells 4 h after infection [ 52 , 130 ]. Single-stranded infectious RNA forms after RIs start to accumulate [ 130 ]. Genomic RNA (plus strand) is the template for complementary minus strand synthesis which in turn is the template for new genomic plus strands that become incorporated into virions [ 131 ].…”
