Replication Banding in two Mediterranean Moray eels: Chromosomal Characterization and Comparison

Abstract: Early and late replication bandings have been obtained by in vitro BrdU incorporation in the Mediterranean Muraenidae species Muraena helena and Gymnothorax unicolor, and used to characterize their karyotypes. A comparative analysis of the banding patterns allowed to point out high karyotype similarity as well as chromosome rearrangements that occurred in karyotype evolution between these species.

“…, 1991;Maistro et al . , 1999], characterization of homeologous chromosome regions [Giles et al, 1988;Salvadori et al, 2003], heterochromatin replication dynamics, and origin and diversification of sex chromosomes [Bertollo et al, 1997;Molina and Galetti, 2007]. Here, the array of late replicating bands along the chromosomes corresponded to the C-positive heterochromatin distribution for both species.…”

“…This failure has been attributed to technical problems in chromosome preparations (Gold et al 1990) or differences in the structure and composition of DNA from fish chromosomes compared to other vertebrates (Medrano et al 1988;Oliveira et al 2002). In spite of what has been mentioned above, karyotype analyses with G and R bands have contributed to comparative analyses between species and chromosome rearrangement detection (Gold et al 1990;Almeida-Toledo et al 1988;Vênere and Galetti 1995;Bertollo et al 1997;Maistro et al 1999;Vasconcelos and Martins-Santos 2000;DanielSilva andAlmeida-Toledo 2001, 2005;Salvadori et al 2003;Boroń 2003).…”

“…Although scarce, karyotypical descriptions using replication bands in fishes have proved to be very elucidative, facilitating the pairing and the observation of homeologies among closely related species (Giles et al 1988;Almeida-Toledo et al 1988;Salvadori et al 2003), supporting a functional and dynamic chromosomal approach. Studies about replication bands in fishes bearing sex chromosomes are unusual (Bertollo et al 1997), and have not been performed in Anostomidae representatives so far.…”
