Repeated rat-forced swim test: Reducing the number of animals to evaluate gradual effects of antidepressants

Abstract: The forced swim test (FST) is a pre-clinical test to short and long term treatment with antidepressant drugs (ADT), which requires between-subject designs. Herein a modified protocol of the FST using within-subject design (repeated rat-FST) was evaluated. Male Wistar rats were submitted to 15 min of swimming (Day 1: pretest) followed by three subsequent 5 min-swimming tests one week apart (Day 2: test, Day 7: retest 1, Day 14: retest 2). To determine the temporal and factorial characteristics of the variables … Show more

“…Because this test can be repeated (Mezadri et al, 2011), we used a cross-over design for treatment, allowing for within-animal repeated-measure analysis. In RC::PDq;Pet1-cre mice, we confirmed the absence of genotype effect upon Veh exposure (F(1,190) = 1.446, p = 0.2366, Figure 2G), and found an effect of treatment (F(1,190) = 7.420 p = 0.0097, Figure 2H).…”

“…However, in earlier studies, the tricyclic antidepressant desipramine, the tetracyclic antidepressants maprotiline and mianserine and the norepinephrine–dopamine reuptake inhibitor nomifensine weren’t effective in rats which had not received the pretest session [162]. Antidepressant effects of fluoxetine treatment were detected at lower than the usual dose with several re-test sessions in the rat FST[49]. …”

“…1). Time between the tests was at least 2 weeks, so as to avoid any previous forced swimming experience to affect an animal’s performance (De Pablo et al, 1989; Mezadri et al, 2011). The test consisted of a single 5-minute-long session in a tank filled with water at 22°-25°C.…”
