“…We also found that YKS did not affect the pentobarbitalinduced sleeping time in group-housed mice, whilst YKS prolonged the pentobarbital-induced sleeping time in socially isolated mice to levels similar to the grouphoused mice, suggesting that YKS attenuates the social isolation stress-induced increase of arousal level. We previously reported that acute treatment with YKS does not affect ambulation or rearing in the open-field test in mice (10). In this study, YKS has a selective effect on pentobarbital-induced sleep in socially isolated mice.…”

Pentobarbital sodium salt was purchased from Tokyo Chemical Industry Co. (Tokyo) and was dissolved in saline.…”

“…The doses of YKS and quetiapine were chosen based on previous reports. [12][13][14] Open-Field Test The procedure for the open-field test followed that previously described by Egashira et al 15) Mice were singly placed in the center of a circular open field (grey iron, diameterϭ60 cm) enclosed by a parapet (height, 50 cm) with an upper opening (diameter, 90 cm). The floor was divided into 19 equal blocks by thin black lines.…”

“…Yokukansan did not increase extrapyramidal symptoms. Yokukansan controls the concentration of serotonin in the synaptic cleft via partial agonist of 5-HT1A receptors (Terawaki et al, 2016), and 5-HT2A receptor expressions are downregulated by yokukansan (Egashira et al, 2008). Yokukansan inhibits the release of glutamate resulting in a decreased concentration of glutamate at synaptic clefts in the brain, and promotes glutamate uptake by astrocytes (Takeda et al, 2008a(Takeda et al, , 2008bKawakami et al, 2010).…”
