Repair and mutagenesis of the genome of a deletion mutant of the coronavirus mouse hepatitis virus by targeted RNA recombination

Abstract: The genetic characterization of a nucleocapsid (N) protein mutant of the coronavirus mouse hepatitis virus (MHIV) is described. The mutant, Albany 4 (Alb4), is both temperature sensitive and thermolabile. Analysis of the progeny of a mixed infection showed that the defective Alb4 allele is recessive to wild type, and its gene product is diffusible. The N protein of Alb4 was found to be smaller than its wild-type counterpart, and sequence analysis of the Alb4 N gene revealed that it contains an internal deletio… Show more

“…The plaque-cloned A59 strain of MHV (MHV-A59) (Lai et al, 1981) and its temperature-sensitive mutant Alb4 (Koetzner et al, 1992) were used for this study. MHV-A59 and Alb4 stock viruses were propagated in mouse DBT cells (Hirano et al, 1974) at 37 • C and 33 • C, respectively.…”

“…We assumed that N proteins in MHV virion nucleocapsid were tightly associated with each other, and asked whether this putative tight N protein association actually was maintained after RNase A-treatment of the virion nucleocapsid had extensively degraded the RNA. MHV-A59 and its temperature-sensitive mutant Alb4 (Koetzner et al, 1992) were used to address this question. MHV N protein consists of three domains, each of which is connected by a short spacer sequence (Koetzner et al, 1992).…”
