Renoprotective effect of Nigella sativa against cisplatin-induced nephrotoxicity and oxidative stress in rat

Abstract: Cisplatin is one of the important antineoplastic drugs. Its clinical use has been restricted due to severe kidney toxicity. Nigella sativa (N. sativa) is an herbaceous plant with many pharmacologic effects. In the present study, we evaluated the protective effects of aqueous-ethanolic extract of N. sativa and Vitamin E on cisplatin-induced nephrotoxicity in rats. Eighty male rats were divided into eight groups: control, cisplatin (6 mg/kg; ip), preventive Vitamin E (100 mg/kg), preventive N. sativa (100,200 mg… Show more

“…Changes in serum creatinine concentration more reliably reflect changes in Glomerular filtration rate (GFR) than do changes in serum urea concentrations (34). The rise in serum level of these chemicals indicates a decline or failure in renal function to filter waste products from the blood and excrete them in the urine (35). In the present study, the sub-acute oral administration of the extracts did not show significant alteration in the serum levels of urea and creatinine concentrations in Mice administered with both doses as compared to the controls.…”

“…According to the experiments that were conducted on humans and animals, the main cause of nephrotoxicity induced by aminoglycosides is oxidative stress (20,21). Oxidative stress occurs when there is an imbalance between oxidants like ROS and antioxidants and is caused by a decrease in antioxidants or when they are depleted as a result of lipid peroxidation, inactivation of antioxidant enzymes, cellular running out of GSH, or destruction of DNA (22).…”

“…A recent study suggested that acute kidney injury is the most serious side effect of cisplatin, an important antitumor agent. Lipid peroxidation and oxygen free radicals are important mediators of cisplatin-induced nephrotoxicity [21]. The molecular role of TGF-1β is critical in driving mechanisms of cellular repair.…”
