Renal kallikrein mRNA localization by in situ hybridization

Abstract: Tissue kallikrein gene expression in rat kidney was examined by in situ hybridization histochemistry. A rat tissue kallikrein cDNA probe, 534 bases in length and complementary to the 3' end of kallikrein mRNA was first used in Northern blot analysis to demonstrate the existence of tissue kallikrein mRNA in rat kidney. Then, kallikrein mRNA's localization in rat kidney sections was studied in situ hybridization histochemistry using the same probe. Positive signals were concentrated in the renal cortex at the va… Show more

“…Kallikrein that produces bradykinin is biosynthesized in kidney cortex (Xiong et al, 1989). Kallikrein 7 (Klk7) is found in Table 5 showing 15-fold and 285-fold expression in medulla and cortex, respectively, compared with papilla.…”

“…The recent finding of kallikrein mRNA in the distal renal tubule and at the vascular pole of the glomerulus suggests that an intrarenal kallikrein-kinin system may be involved in the regulation of blood pressure. 27 Decreased intrarenal activity of the kallikrein-kinin system and attendant decreases in prostaglandin synthesis might give rise to an increase in blood pressure by inducing a shift in the pressure-natriuresis curve.…”

“…Tissue kallikrein mRNA is expressed predominantly in the cells of the distal tubules and also in the vascular pole of the glomeruli (89) and in the CNT of the outer cortex (90). Another report (91) states that kallikrein is present in the granular peripolar cells of the glomeruli of the human kidney, but that its mRNA is not located at this site.…”
