Renal Ischemia/Reperfusion Mitigation via Geraniol: The Role of Nrf-2/HO-1/NQO-1 and TLR2,4/MYD88/NFκB Pathway

Abstract: Background: Renal ischemia/reperfusion injury is a clinically recurrent event during kidney transplantation. Geraniol is a natural monoterpene essential oil component. This study aimed to inspect geraniol’s reno-protective actions against renal I/R injury with further analysis of embedded mechanisms of action through scrutinizing the Nrf-2/HO-1/NQO-1 and TLR2,4/MYD88/NFκB signaling pathways. Methods: Wistar male rats were randomized into five groups: Sham, Sham + geraniol, Renal I/R, and two Renal I/R + gerani… Show more

“…Another experiment showed the aforementioned inhibitory effects were mediated by the inhibition of iNOS and COX2 enzyme induction. The induction of iNOS and COX-2 has also been reported in cyclophosphamide-induced cystitis in rats [39]. In addition, iNOS and COX-2 inhibitors have been shown to reduce hyperactivity.…”

“…The Nrf2 signaling pathway is an important intracellular pathway that plays a key protective role in regulating intracellular oxidative stress and antioxidant defense 67 . Nrf2, a transcription factor, regulates the expression of various antioxidant and detoxification enzymes, including GSH, SOD, and HO-1, to counteract oxidative stress and maintain intracellular oxidative homeostasis 68 . Therefore, higher activation of the Nrf2 signaling pathway results in lower production of inflammatory factors; (3) Activation level of the NFκB signaling pathway: NFκB, a transcription factor, plays a crucial role in cellular signaling and is closely associated with the regulation of inflammatory factors 69 .…”

“…The Nrf 2/ARE signaling pathway is a classical anti-oxidative stress pathway, which is regulated by nuclear factor E2 related factor 2 (Nrf 2) [ 18 , 19 ]. Under physiological conditions, the binding of Nrf 2 to KEAP1 is localized in the cytoplasm, and Nrf 2 is regulated at low protein levels [ 20 ].…”
