The formation of placenta at the beginning of pregnancy and its separation at parturition require not only deep remodelling of extracellular matrix, which mainly consists of proteins conjugated with sugar moieties, but also the cooperation with cells from both maternal and foetal parts of placenta. The aim of the study was to compare the patterns of selected conjugated proteins with sugar moieties between pregnant and term placenta as well as between released and retained placenta in cows. Placental samples from healthy pregnant cows (3–5 months of pregnancy) were collected at a slaughterhouse (n = 6), and parturient samples were collected during caesarean section at term and retrospectively divided into retained (n = 6) and released (n = 6). The pattern of selected sugar moieties conjugated with proteins was detected by use of lectin blotting with Phaseolus Vulgaris leucoagglutinin, Maackia Amurensis and Sambucus Nigra (Elderberry). The comparison and analysis of obtained band patterns showed differences between their number, molecular weight and abundance related to the intensity of staining. Samples from 3 to 4 months showed similarities, while at the 5th month, clear differences were visible in all 3 lectins, which were used in this study. Samples from retained/released placenta expressed significant differences in PHA‐L and SNA pattern in the foetal part. Obtained results indicate that the development of placenta related to extracellular matrix and accompanying cells from both sides of placenta shows dynamic changes during pregnancy. Moreover, in the case of animals with the retention of foetal membranes the patterns of proteins conjugated with sugar moieties are altered, suggesting that the changes in extracellular matrix metabolism can be involved in the attachment and detachment of the placenta in cows.

Reproduction is a critical aspect of livestock production, with mitochondria playing a pivotal role in providing the necessary energy for this high-demanding process. Maternally inherited mitochondria are known to influence various cellular functions, including ATP production and apoptotic regulation, and possess their own genome, including the Displacement loop (D-loop). This study aims to explore the mitochondrial DNA (mtDNA) D-loop SNPs and their potential association with reproductive traits in Gir cattle, an indigenous breed. Two groups of animals were categorized based on their reproductive performance, with good performers demonstrating shorter calving intervals, gestation lengths, days open, and fewer services per conception. Total DNA was isolated from blood samples, and mtDNA D-loop sequences were obtained through Sanger sequencing, resulting in 29 haplotypes. Comparative analysis revealed that animals with poor reproductive performance exhibited a higher number of polymorphic sites, transition/transversion biases, and occurrences of insertions/deletions (INDELs) compared to the good performers. Specifically, good performers showed higher A/G and G/A substitutions, while poor performers exhibited higher T/A, C/A, A/T, G/T, and G/C substitutions. The A/G transition at Position 869 was significantly associated with poor reproductive performance, impacting secondary structure stability and likely affecting energy efficiency. Validation of these findings in a larger Gir cattle population and exploring mtDNA polymorphisms in coding regions may establish a maternal lineage-based biomarker for assessing reproductive efficiency in this indigenous breed. This study sheds light on the significance of mitochondrial genetic factors in shaping reproductive traits and offers insights into sustainable livestock production.

In addition to its many functions, the placenta is characterized by the intensity and characteristics of its hormonal functions. It has been reported that the development and vascularization of the placenta with normal fetal growth is regulated by many factors such as growth factors, transcriptional factors, and homeobox proteins. This study was conducted to determine the distribution and possible physiological roles of HOXA11 and HLX in the uterus and placenta of pregnant cows. In this study, 27 pregnant Holstein breed cow uteri and placentas obtained from private slaughterhouses were used as tissue samples. In order to determine the pregnancy period of the obtained uterus, the ages of the fetuses were calculated according to the age calculation formula. The uteri were classified under three different groups as the first (69-89 days), second (99-178 days), and third (190-269 days) periods of pregnancy so that there are 9 animals in each group. Tissue samples were then subjected to immunohistochemistry to demonstrate the presence of HOXA11 and HLX proteins. It was observed that HOXA11 and HLX immunoreactions occurred prominently in the luminal and glandular epithelial cells of the uterus during pregnancy, and the maternal epithelial cells of the placenta and UTC and TGC/BNCs. In addition, it was determined that HOXA11 immunoreaction also formed in some vascular endothelial cells. Immunohistochemical findings in this study suggested that Hoxa11 and HLX may affect the proliferation and differentiation of cells in the uterus and placenta during pregnancy in cows, and that these proteins may contribute to normal uterine physiology, placentation formation, and continuity of pregnancy in cows.Key words: Hox proteins; HOXA11; HLX; placentaIMUNOLOKALIZACIJA PROTEINOV HOXA11 IN HLX V POSTELJICI KRAVE MED BREJOSTJOIzvleček: Za posteljico so poleg številnih funkcij značilne intenzivnost in karakteristike njenih hormonskih funkcij. Ta študija je bila izvedena z namenom določiti porazdelitev in morebitno fiziološko vlogo protnov HOXA11 in HLX v maternici in posteljici brejih krav. Za vzorce tkiv smo uporabili maternice in posteljice 27 krav pasem Holstein, pridobljenih v zasebnih klavnicah. Obdobje brejosti smo izračunali na podlagi starosti plodov po formuli. Maternice smoglede na obdobje brejosti uvrstili v tri skupine po 9 vzorcev: 1 (69-89 dni), 2 (99-178 dni) in 3 (190-269 dni). Prisotnost proteinoHOXA11 in HLX v vzorcih tkiv smo preverili z imunohistokemijo. Proteini HOXA11 in HLX so bili prisotni predvsem v luminalnihin žleznih epitelijskih celicah maternice ter v epitelijskih celicah maternalne posteljice, UTC in TGC/BNC. Prisotnost HOXA1je bila zaznavna tudi v nekaterih žilnih endotelijskih celicah. Rezultati imunohistokemije v tej študiji kažejo na morebiten vplivproteinov HOXA11 in HLX na proliferacijo in diferenciacijo celic v maternici in posteljici krav med brejostjo in na morebitno vlogoteh proteinov pri normalni fiziologiji maternice, nastanku posteljice in vztrajanju brejosti pri kravah.Ključne besede: proteini Hox; HOXA11; HLX; posteljica
