Remarkable heterogeneity displayed by oval cells in rat and mouse models of stem cell–mediated liver regeneration

Abstract: The experimental protocols used in the investigation of stem cell-mediated liver regeneration in rodents are characterized by activation of the hepatic stem cell compartment in the canals of Hering followed by transit amplification of oval cells and their subsequent differentiation along hepatic lineages. Although the protocols are numerous and often used interchangeably across species, a thorough comparative phenotypic analysis of oval cells in rats and mice using wellestablished and generally acknowledged mo… Show more

“…Higher alpha-fetoprotein (AFP) and delta-like protein expression in the proximal oval cells has already been described. 17,18 Similar zonal distribution could be demonstrated for the expression of other differentiation-related genes. Distal and proximal oval cell groups have been microdissected separately from frozen sections, and the relative gene expression has been analyzed by real-time quantitative RT-PCR.…”

“…In support of this, a recent work has delineated the profound phenotypic heterogeneity displayed by oval cells in different rat and mouse models of oval cell-mediated liver regeneration. 48 It seems reasonable that the same diversity may apply to the regulatory mechanisms operating in these cells. Thus, mouse oval cells isolated from CDE-treated mouse livers express cytokines such as lymphotoxin-␤, interferon-␥, interleukin-6, 49,50 suggesting that they may respond both to autocrine and paracrine cytokine stimulation.…”

“…Hepatoblasts are considered a heterogeneous population with potential hierarchical organization and various degrees of differentiation (113). Therefore, many controversial issues including differentiation status and the repopulation capability of markerpositive hepatoblasts have been raised, even using robust stem cell markers (95,114,115).…”
