Reliability of field, greenhouse and cut‐shoot screening procedures for evaluating susceptibility of Scots pine to Melampsora pinitorqua

Abstract: The reliability of ®eld, greenhouse and cut-shoot screening procedures for the assessment of the susceptibility of Scots pine (Pinus sylvestris) genotypes to Melampsora pinitorqua, the causal agent of twisting rust, was evaluated. Fourteen genotypes evaluated on half-sib progenies after natural infection of 2-year-old plants showed comparable rank for rust susceptibility with those evaluated after controlled inoculation of 1-year-old seedlings in a greenhouse. The dynamic of pycnia production was assessed at 2… Show more

“…Aspen is a far less abundant forest tree in Sweden. It is a primary coloniser and was chemically eradicated in many Swedish forests over a few decades in the mid twentieth century because of its association with Melampsora pinitorqua , a rust that has pine as its alternate host (Quencez et al 2001). The surveys were conducted in all parts of the country, and sampling naturally decreased with remoteness and increased with P. tremula density.…”

“…Populus tremula is often infected by leaf rusts belonging to the genus Melampsora (Quencez et al 2001). These rusts are typically macrocyclic and heteroecious, i.e.…”

“…This is important when developing the mixed-effects model. In this study, all possible combinations were fitted and then the best combination with convergence was selected according to the Akaike information criterion (AIC), Bayesian information criterion (BIC), log-likelihood (Loglik) and the likelihood ratio test (LRT) [66,67]. The AIC, BIC and LRT are defined as follows:…”

“…In vitro bioassays with cut plant material obtained from genetic trials are probably the safest alternative (e.g. Quencez et al, 2001;Fedderwitz et al, 2018).…”

“…However, as the outcome of the molecular interactions between the host and the biotic agent may largely differ between excised material and living plants (Zas et al, 2011), results of in vitro screenings may be not good predictors of what occur in field conditions. Previous fine tuning of the phenotyping protocols is, thus, mandatory (Quencez et al, 2001).…”
