Context: Nasal route of drug administration is preferred more and more for the targeted delivery to the brain in current drug development scenario due to its ease of use, reliability, quick action, and lesser side effects. Those CNS drugs which have limited oral bioavailability due to pharmacokinetic consequences and brain barrier repulsion are getting onto this direction. Objective: Quetiapine fumarate, an analogous to above and an antischizophrenic agent, is tested for its diffusion property with and without lipophilic carrier through sheep nasal membrane. Being a BCS class II' and high permeable candidate, it tends to crossover easily, so made up in a simple dispersion. Materials and methods: To improve its diffusion rate, it was embedded into liposomal dispersion, which has proven that it has advanced efficiency for diffusion. For this, both the formulations were checked and compared for their diffusion profile, as it is an essential property for bioavailability through nasal route. Comparison was made on the basis of % drug diffusion within 6 h, rate, mechanism, profile, and coefficient. Results: Liposomal dispersion has been proved superior with greater percentage diffusion of 32.61 ± 1.70 and very high permeability with a coefficient value of 4.1334 ± 0.7321 (Â 10 À 5 cm/s). Diffusion profile comparison bearing dissimilarity of 18 and similarity of 74 indicated that the diffusion profiles of liposomal dispersions and simple dispersion were similar but not identical. Conclusion: Liposomal diffusion supremacy was further sustained by in vivo, ciliotoxicity, and gamma scintigraphy studies.

Abstract. The present investigation was undertaken to fabricate modified transport fluconazole transdermal spray using ethyl cellulose and Eudragit® RS100 as film-forming polymers. Eudragit® RS100 (X 1 ) and ethyl cellulose (X 2 ) were selected as independent variables in 3 2 full factorial design, whereas drug transport in first hour (Y 1 ) and the time required for 50% drug transport (Y 2 ) were selected as dependent variables. Eutectic blend of camphor and menthol was used as permeation enhancer cum solvent for film-forming polymers. The pH, viscosity, volume of solution delivered upon each actuation, spray angle, ex-in vivo physical evaluation and in vitro drug transport of the formulated products were evaluated. The optimized batch B16 containing 5.25% w/w ethyl cellulose and 10.6% w/w Eudragit® RS100 was formulated by overlapping the contour plots of Y 1 and Y 2 . The pH, viscosity, volume of solution sprayed upon each actuation and spray angle of the batch B16 was 6.3, 52.9 cPs, 0.24 ml and 82.6°r espectively. The film of optimized batch was flexible and dermal-adhesive. The responses Y 1 and Y 2 of batch B16 were 7.91 μg/ml and 347 min respectively. The kinetics of drug transport was best explained by the Korsmeyer and Peppas model. The eutectic mixture consisting of equal parts of camphor and menthol showed improved drug permeation through shed snake skin. Short-term stability study demonstrated insignificant changes in performance characteristics.

In this work a different type of formulation, as disc, containing a selected mucoadhesive polymer, fillers, and binders were investigated for their potential as a mucoadhesive gastroretentive delivery system to deliver famotidine in the stomach. Various types of hydrophilic diluents were evaluated for their swelling and mucoadhesive property and one (polyvinylpyrrolidone, PVP) was selected to combine with the selected mucoadhesive polymer (polyethylene oxide, PEO). Discs with different ratios of PEO and PVP were prepared and evaluated for swelling, dissolution, and mucoadhesion. The swelling property of the discs increased as the concentration of PEO was increased and also did the mucoadhesion. These discs retained their integrity and adherence onto gastric mucosa for more than 10h under in-vitro conditions. The PEO, in combination with PVP, yielded a non-disintegrating type mucoadhesive dosage form which was suitable for gastroretentive applications to achieve the desired release profile of the drug.
