Relative Levels of mRNA Encoding Enamel Proteins in Enamel Organ Epithelia and Odontoblasts

Abstract: Amelogenin, enamelin, sheathlin (ameloblastin/ amelin), enamelysin (MMP-20), and KLK4 (EMSP-1) are the major structural proteins and proteinases in developing tooth enamel. Recently, odontoblasts were reported to express amelogenin, the most abundant enamel protein. In this study, we hypothesized that odontoblasts express all enamel proteins and proteases, and we measured their relative mRNA levels in enamel organ epithelia and odontoblasts associated with porcine secretory- and maturation-stage enamel by RT-P… Show more

“…Amelogenin expression was identified in secretory ameloblasts and stratum intermedium cells at the apical bud of the incisor (21,22). Both maturation stage ameloblasts and stratum intermedium cells associated with maturation and secretion stage ameloblasts are amelogenin-negative (20).…”

“…As molecular probing techniques have become more sensitive, these gene expression pattern profiles are being challenged and/or corrected. In mice, the mRNAs of amelogenin, ameloblastin, and enamelin, in addition to being a product of ameloblast cells, are also apparent in presecretory and secretory odontoblasts (18,(43)(44)(45)(46). The mRNA and protein expression of dentin-matrix protein 1 and Dspp, in addition to odontoblasts, are also apparent in presecretory ameloblasts (14,16,17,47,48).…”

“…Amelogenin message and protein have been detected by various strategies in other tissues, including dental pulp cells [34][35][36][37] , periodontium, bone, brain and other tissues 38,39 . The reader is directed to a recent review for discussion 40 , but major defects have not yet been reported outside of the enamel in either murine models or in humans with AI specific gene mutations.…”
