Relationship of Brain Glutamate Response to D-Cycloserine and Lurasidone to Antidepressant Response in Bipolar Depression: A Pilot Study

Abstract: N-methyl-D-aspartate glutamate-receptor (NMDAR) antagonists such as ketamine have demonstrated efficacy in both major depressive disorder (MDD) and bipolar disorder depression (BP-D). We have previously reported that reduction in Glx (glutamate + glutamine) in the ventromedial prefrontal cortex/anterior cingulate cortex (vmPFC/ACC), measured by proton magnetic resonance spectroscopy (1H MRS) at 3T during a ketamine infusion, mediates the relationship of ketamine dose and blood level to improvement in depressio… Show more

“…Our preclinical studies with mice (71), suggest a comparable mice dose of approximately 100 mg/kg for the 60 mg/kg clinical dose. In this study, D-cycloserine, which acts as an agonist at the Dserine/glycine site of the NMDAR, but a ketamine like antagonist at higher doses (130)(131)(132), also attenuated haloperidol induced bradykinesia at doses up to 30 mg/kg, which likely is in the NMDAR agonist range.…”

“…For instance, in animals, D-cycloserine has been combined with pioglitazone as a possible treatment for a mouse model of chronic orofacial neuropathic pain ( 104 ). In humans, D-cycloserine has been used as part of a combination treatment regimen with lurasidone for bipolar depression or acute suicidal ideation and behavior in bipolar depression ( 105 , 106 ).…”

“…Data preprocessing We combined the data from coil elements using unsuppressed water signal as a reference 19,20 . We removed the residual water signal using an SVD-based method [21][22][23] and performed spectral alignment among the 30 water suppressed FIDs by aligning the 2 nd to the 30 th FIDs to the 1 st one, using a fitting procedure like that of Near et al, using the Lorentzian lineshape function. 24 Evaluation of the temperature averaging -by repeated measurements We used two methods to evaluate the performance of temperature averaging.…”
