Relationship between Vitamin D Binding Protein and Aspirin Resistance in Coronary Ischemic Patients:  A Proteomic Study

Abstract: Our aim was to analyze the plasma proteome in aspirin (acetylsalicylic acid [ASA])-sensitive and ASA-resistant coronary ischemic patients. Plasma from 19 ASA-sensitive and 19 ASA-resistant patients was analyzed. For the proteomic study, two-dimensional electrophoresis was performed. The expression of one isotype of the fibrinogen γ chain and three isotypes of haptoglobin was increased in ASA-resistant patients. Three vitamin D binding protein isotypes were increased in ASA-resistant patients. In vitro incubati… Show more

“…As reported, densitometric analysis of each spot was calculated using albumin as a reference value (Mateos-Cáceres et al 2004). In previous works, we and others have identiWed by mass spectrometry some of the here-reported proteins (Alonso-Orgaz et al 2006;López-Farré et al 2007;Koomen et al 2005;Hlavaty et al 2003). However, in order to conWrm the identity of some of the here-measured plasma proteins, we identiWed again some of the studied plasma proteins by mass spectrometry (data not shown).…”

“…As reported (Mateos-Cáceres et al 2004;Alonso-Orgaz et al 2006;López-Farré et al 2007), 500 g of total plasma proteins, determined by bicinchoninic acid reagent (Pierce), were homogenized in a lysis buVer containing Bio-Lyte TM ampholytes (Bio-Rad). Plasma samples were loaded on immobilized pH gradient IPG strips (pH 4-7), and isoelectric focusing was performed using a Protean IEF cell system (Bio-Rad Labs).…”

“…Interestingly, in this study we observed that the plasma of CHD patients contains greater amounts of Gc-globulin compared with healthy subjects. Previous studies [25] demonstrated that three vitamin D binding protein isotypes were increased in asprin-resistant CHD patients and this could reduce the inhibitory effect of asprin on thromboxane A 2 production. Thus, this may suggest that the increased Gc-globulin level in this study may be associated with a large number of asprin-resistant patients included in our study or with increased reactively due to sudden severe depletion.…”

“…Moreover, differential proteomic analysis has been addressed to elucidate effects on plasma proteome of different pharmacological treatments (AlonsoOrgaz et al, 2006;López-Farré et al, 2007). In this regard, the most applied proteomic methodologies were gel electrophoresis coupled to mass spectrometry analyses followed by gel free proteomic approaches such as LC MS/MS (Donahue et al, 2006;Wilson et al, 2007;Prentice et al, 2010) and microarrays (Tabibiazar et al, 2006).…”
