Relationship between the intrahepatic expression of interferon‐α receptor mRNA and the histological progress of hepatitis C virus‐associated chronic liver diseases

Abstract: Histological progress is one of the predictors of an unfavourable response to interferon (IFN) therapy in hepatitis C virus (HCV)-associated chronic liver diseases (CLD). The aim of the present study was to investigate whether histological progress has an association with the expression of IFN receptor (IFN-Rc) in the liver. Expression of mRNA of the IFN-Rc for IFN-alpha was investigated by reverse transcription polymerase chain reaction using liver biopsy specimens from 37 HCV-associated CLD comprising 11 liv… Show more

“…The method of mRNA measurement may be applicable in other organ disorders, as has been hypothesized and found for instance in the liver (44), the lungs (45), the heart (46), and in cancer research (47,48). In conclusion, the extent of histologic damage is negatively correlated with the GFR at time of biopsy, but it does not predict the rate of change in GFR after taking of the biopsy.…”

“…It is known that the type-I IFN receptor mRNA level in the liver is low in liver cirrhosis [Ishimura et al, 1996;Yatsuhashi et al, 1996;Mizukoshi et al, 1998]. Whether HBV co-infection has an essential role in the development of cirrhosis in chronic hepatitis C, however, is not clear.…”

“…Some studies have found no correlation betw een IFNa /b receptor mRNA expression and histology, (20) and others have found lower IFNAR mRNA expression with "histologic progression" of HCV. (19) The HAI, excluding cirrhosis, is not usually considered a predictor of response to IFN therapy. (9,10) Therefore, although responders to IFN therapy had a higher IFNAR mRNA expression than nonresponders, it is not totally surprising that IFNAR mRNA expression did not correlate with the HAI.…”
