Relationship between seminal plasma tuberoinfundibular peptide of 39 residues and sperm functional attributes in buffalo (Bubalus bubalis)

Abstract: The buffalo seminal plasma protein profile and its relationship with sperm quality have not been studied in detail. Thus, the aim of the present study was to profile buffalo seminal plasma proteins and to assess the relationship between differentially expressed proteins and sperm characteristics. Semen samples (n = 44) were collected from 11 Murrah buffalo bulls (four ejaculates from each animal) and seminal plasma protein profiling was performed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis. Ma… Show more

“…An ejaculate having less percentage of total motile spermatozoa may fail to reach the site of fertilization due to lack of motility regulating factors secreted in epididymis (Fu-Jun & Xiao-Fang, 2012;Yoon et al, 2016) and proteins such as CatSper, IGF1, and ODF1 (Tamburrino et al, 2014;Cabrillana et al, 2017;Selvaraju et al, 2016b) BSP1 (Somashekar et al, 2015), and TIP39 (Selvaraju et al, 2016a). “…In the present study, total sperm motility, percentage of type-A sperm, sperm acrosome reaction, sperm fructose uptake, and sperm intracellular calcium signaling were significantly higher in fertile group than the low fertile and infertile groups. An ejaculate having less percentage of total motile spermatozoa may fail to reach the site of fertilization due to lack of motility regulating factors secreted in epididymis (Fu-Jun & Xiao-Fang, 2012;Yoon et al, 2016) and proteins such as CatSper, IGF1, and ODF1 (Tamburrino et al, 2014;Cabrillana et al, 2017;Selvaraju et al, 2016b) BSP1 (Somashekar et al, 2015), and TIP39 (Selvaraju et al, 2016a). The low percentages of acrosomal reacted spermatozoa of low and infertile groups might suggest the inability of spermatozoa to undergo acrosome exocytosis and lack of calcium-mediated signaling cascades further affects penetration of zona pellucida (Tranter et al, 2000).…”

“…Moreover, since the fertility of thousands of buffaloes is at stake by a single bull, it is therefore imperative to elucidate the buffalo sperm surface pro le and their molecular functions that can help to understand their role vis-à-vis male fertility. Although a handful of studies are available on buffalo sperm proteome, these contemporary studies exhibit an inclination towards either seminal plasma proteins or biomarker discovery [33][34][35][36][37]. The proteomics analysis of semen is considered complicated since it contains secretions from various accessory glands and its proteome varies between and within individuals and exhibits seasonal and temporal uctuations [20,38].…”
