Relationship between miRNA and ferroptosis in tumors

Abstract: Malignant tumor is a major killer that seriously endangers human health. At present, the methods of treating tumors include surgical resection, chemotherapy, radiotherapy and immunotherapy. However, the survival rate of patients is still very low due to the complicated mechanism of tumor occurrence and development and high recurrence rate. Individualized treatment will be the main direction of tumor treatment in the future. Because only by understanding the molecular mechanism of tumor development and differen… Show more

“…This is the case for breast cancer, which was treated with Metformin to target the mir-324-3p/GPX4 axis in a pro-ferroptotic manner [ 110 ]. In addition, Icariside II is a natural active flavonoid that affects this axis and promotes ferroptosis in renal cell carcinoma [ 111 ]. Curcumenol is another natural molecule able to act as a ferroptosis inducer via the lncRNA H19/miR-19b-3p/FTH1 axis.…”

“…In addition to the enhanced glycolysis process, the enhancement of glutamine decomposition is also a characteristic of tumor cells, and GLS is a key enzyme in the glutamine decomposition process. GLS can decompose glutamine (Gln) into glutamic acid (Glu), and then Glu generates α-ketoglutarate (α-KG) under the action of glutamic acid transaminase 1 (GOT1) [ 89 , 90 ]. The α-KG is an important intermediate product of the tricarboxylic acid (TCA) cycle, and the increase of α-KG can promote the TCA cycle to produce more energy, nucleotides, lipids, amino acids and other substances required by cells, which is conducive to cell growth and survival [ 91 93 ].…”

“…First, they have poor specificity and a rather large number of targets. Second, the mechanism of targeting miR-1-3p is also unclear [ 90 ]. Therefore, in order to better target the target molecule, gene drugs will become a hot spot for future research.…”

“…MiRNAs, short single-stranded RNAs, regulate ferroptosis by targeting essential regulators involved in this process, such as GPX4 and acyl-CoA synthetase long-chain family member 4 (ACSL4). Through binding to specific mRNA sequences, miRNAs influence the expression of these proteins ( Wen et al, 2015 ), thereby contributing to the regulation of ferroptosis ( Tabnak et al, 2021 ; Dai et al, 2022 ). Conversely, lncRNAs and circRNAs impact ferroptosis by acting as competitive endogenous RNAs (ceRNAs) or sponges for miRNAs ( Zhou et al, 2022a ; Dabbaghi et al, 2023 ; Tabnak et al, 2023 ).…”
