Background Epidemiological studies indicate that disruption of circadian rhythm by shift work increases the risk of breast and prostate cancer. Our studies demonstrated that carcinogens disrupt the circadian expression of circadian genes (CGs) and circadian-controlled genes (CCGs) during the early stages of rat mammary carcinogenesis. A chemopreventive regimen of methylselenocysteine (MSC) restored the circadian expression of CGs and CCGs, including PERIOD 2 (PER2) and estrogen receptor β (ERS2), to normal. The present study evaluated whether changes in CG and CCG expression in whole blood can serve as indicators of circadian disruption in shift workers. Methods Fifteen shift workers were recruited to a crossover study. Blood samples were drawn before (6 PM) and after (8 AM) completing a night shift after at least 7 days on floating night-shift rotation, and before (8 AM), during (1 PM), and after (6 PM) completing 7 days on day shift. The plasma melatonin level and messenger RNA (mRNA) expression of PER2, nuclear receptor subfamily 1, group d, member 1 (NR1D1), and ERS2 were measured, and the changes in levels of melatonin and gene expression were evaluated with statistical analyses. Results The mRNA expression of PER2 was affected by shift (p = 0.0079); the levels were higher in the evening for the night shift, but higher in the morning for the day shift. Increased PER2 expression (p = 0.034) was observed in the evening on the night versus day shifts. The melatonin level was higher in the morning for both day shifts (p = 0.013) and night shifts (p < 0.0001). Conclusion Changes in the level of PER2 gene expression can serve as a biomarker of disrupted circadian rhythm in blood cells. Therefore, they can be a useful intermediate indicator of efficacy in future MSC-mediated chemoprevention studies.

Objective: Thiol/disulphide homeostasis has a crucial role for life. The aim of this study was to compare thiol/disulphide homeostasis in healthcare professionals working in the operating room (Group 1) and healthcare professionals working in the emergency department (Group 2). Materials-Methods: This was a descriptive study. Fortynine operating room volunteers (Group 1) and 49 emergency department volunteers (Group 2) were included in this study. Thiol and disulphide concentrations were measured using the novel automated measurement method developed by Erel & Neşelioğlu in 2014. Result: The thiol/disulphide levels were compared in both groups; Native thiol in group 1 was 353,57 ± 49,86 μmol.L-1, and it was 333,73 ± 37,38 μmol.L-1 (p = 0.028) in group 2. total thiol values were 384,71 ± 52,36 μmol.L⁻¹ and 370,94 15,56 ± 5,36 μmol.L-1(P =0,133), respectively in group 1 and 2. In group 1,disulphide levels were 15,56±5,36 μmol.L-1, in group 2 it was 18,61 ± 7,71 μmol.L-1 (p = 0,025), disulphide/native thiol in group 1 was 4,47 ± 1 %, and in group 2 it was measured %, 5,73 ± 2,95% (p = 0.009). Disulphide/total thiol in the group 1 was 4,06 ± 1,32% and 5,03 ± 2,13% (p = 0,008) in group 2. Native thiol / total thiol levels were 90,23 ± 12,16% in group 1 and 89,93 ± 4,26% in group 2 (p = 0,871). Native thiol level in group 1 and high level of disulphide in group 2 indicate that the exposure to oxidative stress in the emergency department is higher than in the operating room. Conclusion: Our results show that participants of emergency room group have statistically higher levels of disulphide than participants working in operating room which may suggest presence of oxidative stress related with workplace exposures. RESUMO: Este estudo tem como objetivo identificar a influência das cargas de trabalho na saúde do trabalhador de enfermagem. A busca foi realizada em novembro de 2016, em quatro bases de dados, estabelecendo-se critérios de inclusão e exclusão, sendo selecionados 50 estudos. Os resultados destacam os conceitos, tipos e condições de trabalho que predispõem as cargas de trabalho, evidenciando as consequências na saúde da equipe de enfermagem por meio dos adoecimentos, que acarretam afastamentos e absenteísmo, estresse, desgastes físicos e emocionais, riscos para acidentes de trabalho, entre outros. Conclui-se que a identificação dessas influências possibilita a implementação de ações organizacionais, de prevenção de acidentes e doenças relacionadas ao trabalho. Se destacan los conceptos, tipos y condiciones de trabajo que predisponen las cargas de trabajo, evidenciando las consecuencias en la salud del equipo de enfermería por medio de las enfermedades, las cuales resultan alejamiento y absentismo, estrés, desgastes físicos y emocionales, riesgos para accidentes de trabajo, entre otros. Se constata que la identificación de esas influencias posibilita la implementación de acciones organizacionales, de prevención de accidentes y enfermedades referentes al trabajo.
