Acute cold exposure (ACE) stimulates metabolism but data evaluating inflammatory and lipid biomarkers related to cardiovascular disease in response to ACE are lacking. Therefore, we investigated the relationship between ACE and inflammatory and lipid biomarkers. Methods: Twenty subjects underwent 30 min of ACE with blood drawn: 1) pre-ACE (baseline), 2) at 30 min ACE, and 3) 2 h post-ACE. Plasma was analyzed for 10 cytokines (monocyte chemoattractant protein-1 (CCL2), interleukin (IL)-1β, IL-2, IL-4, IL-6, IL-8, IL-10, granulocyte-macrophage colony stimulating factor (GM-CSF), tumor necrosis factor-alpha (TNF-α), interferon-gamma (INFγ)), 9 acute phase proteins (ferritin, fibrinogen, procalcitonin, serum amyloid A, serum amyloid P, tissue plasminogen, C-reactive protein, haptoglobin, α-2-macroglobin), and 5 lipid biomarkers (triglycerides, nonesterified fatty acids (NEFA), low-and high-density lipoprotein (LDL, HDL), and total cholesterol). Heart rate, whole body oxygen consumption (VO2), energy expenditure (EE), shivering sensations, and pain were measured prior and during ACE. Results: ACE increased heart rate by 11%, shivering sensations by 4-fold, pain by 2-fold, VO2 by 50%, and EE by 52%. IL-1β increased after 30 min ACE by 24% (p=0.048) and 2 h post ACE by 65% (p=0.01). Alpha-2-macroglobin increased after 30 min ACE by 16% (p=0.005) and returned to baseline levels 2 h post-ACE. HDL increased at 2 h post-ACE by 15% (p=0.034). Sex differences were noted between IL-2, IL-6, IL-8, ferritin, α-2-macroglobin and HDL. Conclusions: These findings indicate ACE increases EE and modifies inflammation, the acute phase response, and lipid metabolism.

The study subjects were 192 overweight and obese Thais (BMI > 25.00 kg/m2), and 103 Thai controls (BMI = 18.50-24.99 kg/m2), whose ages ranged from 18-60 years. All subjects were evaluated for serum Alpha-2-macroglobulin (A2M), globulin, albumin concentration, and polymorphic variation in the A2M gene. Serum A2M and albumin were significantly lower in the overweight/obese group (P < 0.05). For the overweight/obese and control group, the median ages were 38 and 37 years, serum A2M 200.2; 252.0 (mg/L), albumin 4.4; 4.5 (g/dL), and globulin 3.0; 2.95 (g/dL), respectively. A2M deletion polymorphism genotyping showed no association between A2M deletion polymorphism and the two groupings. At serum A2M concentration < 250 mg/L, there was no relationship between A2M deletion polymorphism and age. Serum A2M had a significant negative correlation with age in all subjects (R = 0.09, P < 0.05). The results did not support the hypothesis that A2M deletion polymorphism is associated with a low A2M concentration in overweight/obese subjects. However, serum A2M had a significant negative correlation with age; serum A2M can possibly be used to indicate the aging of cells in vivo, including the brain. Further studies are needed to investigate other A2M genes located on chromosome 12 to prove A2M gene polymorphism's association with low serum A2M and aging.

Jacqueline Pontes Monteiro declaro para os devidos fins que CAROLINA DE ALMEIDA COELHO LANDELL, aluna de Doutorado regularmente matriculada no Programa de Pós-Graduação Saúde da Criança e do Adolescente, sob minha orientação, está apta à defesa pública de sua tese Proteômica em dois grupos metabólicos de crianças e adolescentes com diferente perfil lipídico e glicídico submetidos à suplementação de micronutrientes, que está sendo depositada na presente data. A presente tese teve como objetivo descrever dois diferentes grupos metabólicos em um estudo de intervenção nutricional; descrever o perfil lipídico, os níveis de glicose e os dados proteômicos ao longo do estudo nesses dois grupos. A técnica estatística "k-means clustering" foi utilizada para alocar os participantes em dois grupos metabólicos distintos (cluster 1 e cluster 2), de acordo com os perfis glicídicos e lipídicos (glicemia, triglicerídeos, colesterol total, LDL, HDL e VLDL colesterol) que os indivíduos apresentaram nos três momentos do estudo. O cluster 1 (n = 111) teve melhor perfil glicolipídico e também apresentou menores valores para índice de massa corporal, circunferência de cintura e porcentagem de gordura corporal nos três momentos do estudo, comparado ao cluster 2 (n = 25). A ingestão alimentar não diferiu entre os clusters em nenhum momento do estudo. Com a suplementação de micronutrientes, o cluster 1 apresentou redução de glicemia, LDL e colesterol total, além de diminuir sua ingestão de energia, carboidrato, lipídeo e proteína, enquanto o cluster 2 reduziu LDL, colesterol total e HDL e não alterou sua ingestão de energia e macronutrientes. Foi identificada a expressão de 20 proteínas no plasma dos indivíduos dos clusters 1 e 2, sendo que a maioria delas evoluiu de maneira diferente entre os dois grupos após a intervenção. Com a suplementação, o cluster 1 apresentou aumento na expressão de alpha-1-acid glycoprotein 1, alpha-2-HS-glycoprotein, ceruloplasmin e de fibrinogen alpha, beta e gamma chain, bem como redução na expressão de apolipoprotein A-IV, haptoglobin, Ig alpha-1 chain C region, serotransferrin e vitamin D-binding protein. No mesmo período, o cluster 2 mostrou aumento de alpha-1-antitrypsin, ceruloplasmin, haptoglobin, Ig alpha-1 chain C region e plasma protease C1 inhibitor, além de diminuição na expressão de alpha-1-acid glycoprotein 1 e de fibrinogen alpha, beta e gamma chain. É possível que tenha ocorrido aumento de expressão de proteínas que podem ter auxiliado na melhora do perfil glico-lipídico dos participantes. O cluster de pior perfil parece ter se beneficiado mais com a intervenção em relação à expressão de proteínas do que o cluster de melhor perfil.
