Relationship between acrosome integrity changes and in vitro fertilising ability of bovine spermatozoa

Abstract: This study was designed to investigate the characteristics of acrosomal changes during capacitation of bovine spermatozoa in relationship to in vitro fertility of individual bulls. Motile spermatozoa were separated from frozen-thawed semen by a swim-up procedure and capacitated in IVF-TALP medium with or without heparin. The spermatozoa were evaluated in terms of acrosomal changes at 0, 3, 4, 5, 6 and 8 h of capacitation. Proportions of acrosome-reacted spermatozoa at 5 h and 0 h of capacitation were used for … Show more

“…The findings in this study were in accordance with the report of [15] on Red Sokoto bucks, whose epididymal sperm livability declined in a storage-time dependent manner. The success of assisted reproductive techniques such as artificial insemination (AI) and in vitro fertilization (IVF) depends on the use of a sufficient number of motile acrosome intact sperm that has the potential of fertilizing the oocyte [35]. An intact acrosome prevents the loss of acrosomal enzymes such as hyaluronidase until they are released during acrosome reaction in the female reproductive tract prior to fertilization [36].…”

“…During the fertilisation process, biophysical, biochemical, molecular and metabolic changes occur. https://doi.org/10.17221/48/2022-VETMED Some capacitating agents are secreted in the female reproductive tract during in vivo fertilisation (Maitan et al 2022), but need to add these agents in the in vitro fertilisation (IVF) medium during in vitro conditions (Reckova et al 2015). These agents, heparin, caffeine, calcium ionophore and PHE (d-penicillin, hypotaurine, and epinephrine) have been used singly or combined with others (Waheed et al 2016;El-Ruby et al 2017).…”
