Relation of Polyamine Synthesis and Titer to Aging and Senescence in Oat Leaves

Abstract: Polyamine biosynthesis in senescing leaves of A vena sativa L. was measured by determining the activities of arginine decarboxylase (EC 4.1.1.19), ornithine decarboxylase (EC 4.1.1.17) and S-adenosyl-L-methionine decarboxylase (EC 4.1.1.50). Polyamine content was also estimated by thin layer chromatography and high performance liquid chromatography. Arginine decarboxylase activity decreases progressively in aging attached first leaves and in senescing excised leaves in the dark. Conversely, it increases during… Show more

“…For measurements of protease activity, leaves were sterilized as described earlier (19). The lower epidermis was peeled off and 45-mm-long leaf segments were rinsed in distilled H20 and floated in Petri dishes, stripped side down, on 5 ml of 1 mm phosphate buffer (pH 5.7).…”

“…PAs were extracted from leaf segments (19) with 5% HC104, dansylated and separated on silica gel thin-layer plates, and quantified as described in earlier reports (11,19 (Table I). Dap titer increased 44-fold with 0.5 mm Spd and 66-fold with 1.0 mm Spd during 48-h treatments, while a 3-h exposure to 1.0 mm Spd increased the titer by only 9-fold.…”

“…a-difluromethylarginine inhibited ethylene induced putrescine accumulation and coleoptile elongation. It seems that putrescine biosynthesis might be involved in the ethylene-induced elongation of rice coleoptiles.Polyamines, present ubiquitiously in a wide range of higher plants (8), are proposed to play important roles in the regulation of plant growth and development, including cell division, morphorgenesis, flower initiation, pollen tube growth and senescence (5,8,9,15). Stresses such as oxygen deficiency (19-21), chilling (11), high saline, low pH, and K+ and Mg2+ deficiency (5,8,9) increase polyamine contents and their biosynthetic enzyme activities (5,8,9).…”

“…It was worrisome that the activities of the two enzymes reported for comparable leaves of oat cv Victory seedlings, grown under controlled conditions, were much different from ours and at the same time showed significant variability. The reported ADC and ODC activities varied from 2.2 to about 32 and from 0.8 to 18 nmol/h x mg protein, respectively, with ODC/ADC ratios ranging from 0.1 to 6.0 (16,19,31,33,34). Thus, one might suppose that there is significant intraspecific variation in activity levels and/or substantial dependence on external uncontrolled factors; e.g.…”
