Klebsiella aerogenes strain W70 has separate inducible pathways for the degradation of the pentitols ribitol and D-arabitol. These pathways are closely linked genetically as determined by transduction with phage PW52. There are two regulatory sites for the ribitol catabolic pathway as defined by loci for mutations to constitutive synthesis of ribitol dehydrogenase and D-ribulokinase, rbtB and rbtC. The two control sites are separated by a site represented by the dalB22 mutation. This mutation deprives the cell of the ability to induce synthesis of D-arabitol dehydrogenase and D-xylulokinase activities. Two additional regulatory mutations for the D-arabitol pathway, dalC31 and dalC37, map to the opposite side of rbtB13 relative to dalB22. The order of the genetic sites on August 1, 2020 by guest

1. Coumarin when fed to rabbits is hydroxylated to 3-, 7-and 8-hydroxycoumarins, which are excreted in conjugation. This also occurs in the ferret, guinea pig, mouse and rat, the first three of which also excrete 5-hydroxycoumarin. However, not more than 25 % of the coumarin fed (dose 0 1 g./kg.) can be accounted for as conjugated glucuronic acid and ethereal sulphates excreted in the urine. 2. The ethereal sulphate and glucuronide of 3hydroxycoumarin were isolated from the urine of rabbits fed on coumarin. 7-Hydroxycoumarin was also isolated after acid hydrolysis of the urine. 5and 8-Hydroxycoumarins were not isolated but were detected in the hydrolysed urine by paper chromatography. The hydroxylation of coumarin has also been carried out with Fenton's reagent and an ascorbic acid-iron system. The results, which were not exactly the same as those found in vivo, have been discussed. 3. o-Coumaric acid appears to be excreted unchanged to a large extent by rabbits. It is, however, directly conjugated to a small extent since its ether glucuronide, o-glucosidurono-tran8-cinnamic acid, was isolated and its glycine conjugate, o-hydroxy-tran8-cinnamoylglycine, was detected by paper chromatography in the urine. 4. o-Coumaric acid % also cyclized in vivo, since small amounts of the glucuronide of 4-hydroxycoumarin were isolated from the urine and 7hydroxycoumarin was detected chromatographically. Melilotic acid, the corresponding saturated acid (dihydrocoumaric acid), also yields 4-and 7hydroxycoumarin in the rabbit. 5. The mechanism of the cyclization of o-coumaric acid has been discussed and it is suggested that the coumarin derivatives formed arise from ,-oxidation products of o-coumaric acid. SUMMARY: Five mutants of Escherichia coli which grow to a greater or less extent when the minimal medium is supplemented with vitamin B, have been studied. They fall into two groups according to the further effect of serine + glycine on growth.(1) Two strains showed full and rapid growth with pyridoxine alone and serine + glycine neither replaced this vitamin nor enhanced growth in its presence. (2) Three strains showed delayed and suboptimal growth with pyridoxine alone. With these serine + glycine had two effects: (a) it decreased the lag and enhanced the final extent of growth when added with pyridoxine ; (b) it permitted suboptimal growth in the absence of pyridoxine. The second effect was dependent on (two strains), or greatly increased by (one strain), the concurrent addition of glycolaldehyde.Nothing is known of the pathway of synthesis of vitamin B, by organisms not requiring an external source of it for growth. Mutant strains of Escherichia coli which multiply only when vitamin B, is supplied are more rare than those which respond to vitamin B, or serine or glycine. The simplest explanations of the apparent replacement of the vitamin by these amino acids are: ( a ) the amino acids are end products of reactions catalysed by vitamin B, derivatives; consequently, when provided preformed, they decrease or abolish the need for the vitamin; ( b ) the synthesis of serine and glycine by the organism is limited, and one or other is a reactant a t some step in the biosynthesis of vitamin B,.A detailed study was made of the properties of mutants of this type in an attempt to discover an immediate biochemical lesion resulting from the mutation and possibly to obtain some conception of the route of synthesis of vitamin B, and its derivatives by normal strains of Escherichia coli. METHODSEscherichia coli. Stock cultures were maintained on tryptic meat agar slopes and were subcultured monthly (16 hr. a t 37" , 1955) which required vitamin B, for growth a t 37" but responded also to certain mixtures of amino acids at lower temperatures.Strain B166 was used for most of the work to be described.
