Regulatory Mechanisms of Epigenetic miRNA Relationships in Human Cancer and Potential as Therapeutic Targets

Abstract: Initiation and progression of cancer are under both genetic and epigenetic regulation. Epigenetic modifications including alterations in DNA methylation, RNA and histone modifications can lead to microRNA (miRNA) gene dysregulation and malignant cellular transformation and are hereditary and reversible. miRNAs are small non-coding RNAs which regulate the expression of specific target genes through degradation or inhibition of translation of the target mRNA. miRNAs can target epigenetic modifier enzymes involve… Show more

“…Normal miRNA expression and function can be deregulated in cancer [ 11 , 12 ]. Their critical roles in cancer find evidence in a complex map of interactions underlying the relationship between miRNA regulation and the hallmarks of cancer [ 13 ]. A single miRNA can target multiple mRNAs and one mRNA can also be targeted by multiple miRNAs [ 14 ].…”

“…These four miRNA are all HBx epigenetically dysregulated in various HBV-HCC pathways, as well as exercise diverse roles in both the innate and adaptive immune pathways. In many cases, the same miRNA plays a significant regulatory role in many other cancers like those of the breast, lung and colon ( 147 ). It is important to highlight that the illustrated hypothetical pathways in Figures 2 – 5 occur in a dynamic context and that the degree of influence of any single path is non constant.…”

“…In HBV-HCC and many other cancers, the tumor suppressor miR-101 is regarded as a key miRNA in epigenetic systems ( 147 ) (see Figure 2 ). This HBx-dysregulated miRNA involves both upstream and downstream epigenetic systems and has been widely reported as downregulated in HBV-HCC ( 147 ).…”

“…miRNAs may participate in the epigenetic silencing of genes by DNA methylation, either indirectly through binding to mRNAs encoding transcription factors or by directly affecting the mRNAs coding for DNA methyltransferases [ 13 ]. In turn, the activity of promoters of some miRNAs depends on the expression level of DNA methyltransferases, which form regulatory feedback loops [ 14 ]. According to the base miRWalk, miRanda, RNA22, and Targetscan, one miRNA molecule regulates the expression level of tens to hundreds of genes, and at the same time, one gene is a target for many miRNAs.…”
