Regulators of Cell Division in Plant Tissues. XXII* Physiological Aspects of Cytokinin-induced Radish Cotyledon Growth

Abstract: The cytokinin 6-benzylaminopurine (BAP) markedly stimulated the lateral expansion of excised immature radish cotyledons after a lag period of about 10 h. This growth occurred principally by cell enlargement, especially in the light which enhanced the response. However, a marked response 'to cytokinin occurred in the complete absence of red light during germination, cotyledon excision and incubation. Contact with BAP for 5 h significantly stimulated growth, but a maximum response required more than 24 h of cont… Show more

“…8C). TTiese results are consistent with those seen in the growth of detached radish cotyledons in the light (Gordon and Letham 1975) but not with those seen in detached watennelon cotyledons in the dark or in the light (Longo et al 1978, Lampugnani et al 1980). However, tbe copresence of KC!…”

“…The tRNA gave rise to 700 ng <-/.v[9RlZ. Hence, the amount of tRNA that must be present in sap to give rise to the detected concenttations of c;.v[9RlZ of up to 0-9ng cm "* must be at least 13 fjg cm \ Estimation of total RNA in sap, based on the UV absorption of an RNA nucleotide ftaction prepared according to Gordon & Lethatn (1975), indicated that the total RNA level was <IO ;Ug cm ' in the oat and wheat sap. The level of tRNA would probably be much less.…”

“…Thus, we must determine whether hormones promote growth because they enhance production or absorption of osmotically active solutes or cause wall loosening, or both. For cytokinins, insufficient data are available to decide, even for expansion of excised cotyledons in which the dry weight remains constant during typical growth experiments and cytokinesis is a minor factor (2,7,15).…”
