Regulator of G Protein Signaling Protein 12 (Rgs12) Controls Mouse Osteoblast Differentiation via Calcium Channel/Oscillation and Gαi-ERK Signaling

Abstract: Bone homeostasis intimately relies on the balance between osteoblasts (OBs) and osteoclasts (OCs). Our previous studies have revealed that regulator of G protein signaling protein 12 (Rgs12), the largest protein in the Rgs super family, is essential for osteoclastogenesis from hematopoietic cells and OC precursors. However, how Rgs12 regulates OB differentiation and function is still unknown. To understand that, we generated an OB‐targeted Rgs12 conditional knockout (CKO) mice model by crossing Rgs12fl/fl mice… Show more

“…We found that serum total bile acid and FGF19 levels were positively associated with the BMD of the lumbar spine and hip. Zhenxi Li et al have found that the administration of SH-479, which is a bile acid receptor agonist, to mice with postmenopausal osteoporosis increases BMD and improves bone microstructure (8). Moreover, the use of bile acid was found to be an effective treatment for type 2 diabetes, hyperlipidemia, obesity, and other diseases.…”

“…Moreover, it transduces signals mainly by activating the nuclear receptor, farnesoid X receptor (FXR), membrane receptor, and G protein-coupled bile acid receptor 5 (TGR5) (7). A previous study has revealed the occurrence of rapid and severe bone loss after ovariectomy in FXR knockout mice (8). FXR gene knockout has bidirectional effects on the bones.…”

“…*P < 0.05, **P < 0.01, ***P < 0.005, compared with the controls cellular signaling pathways, such as activation of Smad1/ 5/8 phosphorylation or β-catenin signaling, which can promote osteogenesis 39 . In addition, the activation of ERK is critical for osteoblast differentiation and survival 40,41 . However, the phosphorylation of ERK was shown to be suppressed in osteoblasts similarly to osteoclasts.…”

“…In addition, both extracellular calcium and 1,25(OH) 2 D raise the level of intracellular free calcium as a necessary step toward stimulating keratinocyte differentiation 15 . Accumulating studies have shown that calcium oscillations via restraints of major Ca 2+ entry sources (extracellular Ca 2+ influx and intracellular Ca 2+ release from the endoplasmic reticulum) is required for osteoblast differentiation 31 .…”
