Regulation of Vascular Endothelial Growth Factor Synthesis and Release by Human Luteal Cellsin Vitro

Abstract: Our results suggest that local ovarian factors, rather than hCG, predominate in regulating VEGF mRNA and protein production by human midluteal phase luteal cells. For VEGF, a lack of a direct luteal steroidogenic effect was also demonstrated.

“…These unexpected data could be partially explained by our observation of an Es inhibitory effect on luteal P production. Indeed we previously demonstrated the ability of P to decrease VEGF luteal secretion and expression (13). Based on these data it is tempting to speculate that after an Es-mediated P decrease, the lack of any P inhibitory effect on VEGF production could contribute to increase luteal VEGF release.…”

“…To analyze VEGF messenger RNA (mRNA) modulation in human LC, attention has been focused on diffusible mRNA expression of the VEGF 121 and VEGF 165 isoforms (13). The latter was evaluated by real-time RT-PCR using the i-Cycler iQ system (Bio-Rad Laboratories, Hercules, CA), as previously described (22).…”

“…VEGF is considered to be critical in the regulation of normal and abnormal angiogenesis in the ovary (3,9,(13)(14)(15)(16)(17)25,30), particularly in the newly formed corpus luteum. In primates, VEGF protein is localized in the hormone-producing cells of the corpus luteum, and is highest in the granulosa-derived cells (14)(15)(16)(30)(31)(32)(33)(34)(35). VEGF is fundamental in the physiological angiogenesis and vascularization of the follicular luteinizing granulosa layer during corpus luteum formation (15,16,30,32).…”
