Regulation of the sacPA operon of Bacillus subtilis: identification of phosphotransferase system components involved in SacT activity

Abstract: The sacT gene which controls the sacPA operon of BaciUlus subtilis encodes a polypeptide homologous to the B. subtilis SacY and the Escherichia coli BglG antiterminators. Expression of the sacT gene is shown to be constitutive. The DNA sequence upstream from sacP contains a palindromic sequence which functions as a transcriptional terminator. We have previously proposed that SacT acts as a transcriptional antiterminator, allowing transcription of the sacPA operon. In strains containing mutations inactivating p… Show more

“…4; compare lanes 6 -27 Ϫ/ϩbglF, respectively, with lanes 4 and 5). No phosphorylation by HPr could be detected for the proteins with exchanges C76R, S97P, H101L, E153V, and H160R (lanes 6,12,20,24,26), and signals were extremely reduced with the mutants carrying exchanges of I80 and D100 (lanes 8, 14,16,18). Surprisingly, when cells expressed the Q91R protein, an additional phosphorylated band appeared approximately 3 kDa above the phosphorylated GalK-BglG fusion protein (lanes 10 and 11).…”

“…In addition, the sacPA and the bglPH operons are subject to catabolite repression. The antiterminators controlling the expression of these operons depend on a stimulation by HPr to be active, in addition to negative control exerted by the specific EII (Arnaud et al, 1992;Le Coq et al, 1995). The levanase operon, which encodes a fructosespecific PTS (lev-PTS) and an extracellular enzyme, levanase, is positively regulated by a NifA-like activator protein, LevR.…”

“…Some antiterminators from Gram-positive bacteria were shown to be activated by HPr-catalyzed phosphorylation on one or more of the conserved histidines (13,15,16). The HPr-catalyzed phosphorylation was proposed to be part of a mechanism of carbon catabolite repression (CCR) that operates in Gram-positive bacteria (reviewed in ref.…”
