Regulation of Steroidogenesis and the Steroidogenic Acute Regulatory Protein by a Member of the cAMP Response-Element Binding Protein Family

Abstract: The mitochondrial phosphoprotein, the steroidogenic acute regulatory (StAR) protein, is an essential component in the regulation of steroid biosynthesis in adrenal and gonadal cells through cAMP-dependent pathways. In many cases transcriptional induction by cAMP is mediated through the interaction of a cAMP response-element binding protein (CREB) family member with a consensus cAMP response element (CRE; 5'-TGACGTCA-3') found in the promoter of target genes. The present investigation was carried out to determi… Show more

“…The evidence that ICER acts as a tumor suppressor gene in leukemia cell line HL60, as reported previously for other solid tumors, [28][29][30] first comes from the decreased clonogenicity ability of HL60 þ ICER cells in vitro. We used immunodeficent mice to confirm these data and reveal that HL60 þ ICER-injected mice have a mild leukemia phenotype for the reduced dissemination of tumor in extramedullary sites.…”

“…The 5 0 -flanking -966/-1 bp region of the mouse StAR gene (in relation to the transcription start site) was cloned into the pGL3 basic vector (Promega), upstream of a luciferase reporter gene, utilizing the XhoI and HindIII sites (Manna et al 2002b. The identity of the inserted fragment was tested by restriction endonuclease digestion and by automated sequencing with a Perkin-Elmer Biosystem 310 Genetic Analyzer (ABI PRISM 310; Perkin-Elmer, Norwalk, CT, USA).…”

“…The identity of the inserted fragment was tested by restriction endonuclease digestion and by automated sequencing with a Perkin-Elmer Biosystem 310 Genetic Analyzer (ABI PRISM 310; Perkin-Elmer, Norwalk, CT, USA). Expression plasmids for wild-type CREB (WT-CREB) and non-phosphorylatable CREB (CREB-M1, Ser 133-Ala) have been previously described (Manna et al 2002b). The plasmid (pRL-SV40) containing the Renilla luciferase gene driven by the SV40 promoter (Promega) was used to normalize the transfection efficiency.…”

“…NR4A orphan receptors are transcription factors that belong to the superfamily of steroid nuclear hormone receptors that have been associated with different cellular processes, including inflammation (Murphy et al 2001;Pei et al 2005), steroidogenesis (Manna et al 2002), apoptosis (Winoto and Littman 2002), development of dopaminergic Concise summary Non-selective AR engagement triggers robust induction NR4A orphan receptors in mast cells. Induction of these early responsive genes can be blocked by selective activation of the anti-inflammatory A 2A AR not only in the context of adenosine but also of PMA signaling, suggesting a general mechanism of mast cell regulation.…”
