Regulation of Phagosomal Acidification

Abstract: Vacuolar-type (V) ATPases are thought to be the main determinant of phagosomal acidification. In phagosomes containing mycobacteria, which ostensibly impair the delivery of V-ATPases to the phagosomal membrane, the pH would be expected to be near neutral. This prediction was tested by microfluorescence ratio imaging using macrophages from mice susceptible to mycobacterial infection. Although less acidic than their counterparts containing dead bacteria, phagosomes containing live Mycobacteria bovis were nearly … Show more

“…Thus, phagosomes containing live mycobacteria do not fuse with lysosomes nor do they acidify fully, yet acquire LAMP1 (15,16,61). To ensure that bona fide phagolysosomes are formed in the absence of ⑀-COP we measured phagosomal pH in FcR-ldl cells.…”

“…Acidification of phagosomes has been shown to occur by fusion with late endosomes and lysosomes in mammalian cells (52). We examined by using LysoTracker Red, a membranediffusible probe accumulated in acidic organelles (53), whether the PPV and phagosomes of the amoeba are acidified during maturation.…”
