Regulation of lipoprotein lipase mRNA content in 3T3-L1 cells by tumour necrosis factor

Abstract: Tumour necrosis factor (TNF) was previously shown to suppress lipoprotein lipase (LPL) synthesis and activity in 3T3-L1 adipocytes. The present study examined the effect of TNF on amounts of mRNA for LPL in 3T3-L1 cells. Northern-blot analysis of polyadenylated RNA using a cDNA probe to guinea-pig LPL identified two predominant species of LPL message, 3.7 and 3.9 kilobases in size. The steady-state amounts of these mRNAs increased 10-fold upon expression of the adipocyte phenotype. A single dose of 1.5 nM-TNF … Show more

“…Because TNF-␣ has been previously implicated in the regulation of adipogenesis, insulin resistance, and leptin production, 10,20,21,24,25,33 we sequenced the TNF-␣ gene in SHR and SHR.1N. A total of 26 sequence differences in the TNF-␣ gene were identified between SHR and SHR.1N ( Figure 6).…”

“…Thus, TNF-␣ strongly inhibits adipose conversion and even causes a dramatic dedifferentiation of adipocytes in culture (3)(4)(5)(6). Moreover, it is also documented that TNF-␣ decreases the synthesis and activity of several proteins essential for lipogenesis and triglyceride accumulation in adipocytes.…”

“…Moreover, it is also documented that TNF-␣ decreases the synthesis and activity of several proteins essential for lipogenesis and triglyceride accumulation in adipocytes. These include lipoprotein lipase (5,(7)(8)(9), acetyl-coenzyme A carboxylase (4,10,11), acyl-coenzyme A synthetase (12), stearoyl-coenzyme A desaturase (12), and the insulin-sensitive glucose transporter GLUT4 (13,14).…”

“…Treatment of preadipocytes with TNFα inhibits adipogenic conversion [71,81] and treatment of adipocytes results in what has been termed "dedifferentiation" [70,71,75]. Exposure of adipocytes to TNF α has long been known to promote adipocyte lipolysis [66,70]; this can be partly attributed to TNFα-mediated inhibition of LPL activity, which was first noted two decades ago [1,8,51]. The dedifferentiated phenotype that results from TNFα treatment has been ascribed, at least in part, to the TNFα-mediated transcriptional downregulation of the master adipocyte transcription factor PPARγ and thus the subsequent loss of PPARγ-mediated transactivation of select adipocyte genes [81].…”
