Regulation of innate immune signaling pathways by autophagy in dengue virus infection

Abstract: Autophagy is not only an intracellular recycling degradation system that maintains cellular homeostasis but is also a component of innate immunity that contributes to host defense against viral infection. The viral components as well as viral particles trapped in autophagosomes can be delivered to lysosomes for degradation. Abundant evidence indicates that dengue virus (DENV) has evolved the potent ability to hijack or subvert autophagy process for escaping host immunity and promoting viral replication. “…However, several forms of selective autophagy have been identified which lead to the degradation of specific organelle, proteins, and pathogens in mammals 22 . The canonical function of autophagy, that is, the formation of double‐membrane autophagosomes that deliver cargo to endolysosomal compartments, may exert several different antiviral roles, including (1) the degradation of intracytoplasmic viral components 23 ; (2) the activation of innate immune signaling 24 ; (3) the activation of adaptive immunity by the presentation of endogenous viral antigens to major histocompatibility complex (MHC) class I and MHC class II molecules 25 ; (4) the regulation of innate immune activation by mitochondrial quality control 26 ; and (5) the promotion of cell survival 27 . Autophagy acts as a double‐edged sword in viral infections.…”

“…Activation of FOXO3a can suppress pro-inflammatory cytokines and reduce inflammation, thereby preventing tissue damage and improving clinical outcomes. Moreover, FOXO3a has been shown to promote autophagy which helps in clearing viral infections by degrading and recycling viral components (Wan et al, 2022). Therefore, the identification of genes involved in the FOXO signaling pathway may provide a novel therapeutic approach for COVID-19 and HFRS by regulating immune responses, oxidative stress, and inflammation.…”

“…Autophagy is an evolutionarily conserved cellular process induced by nutrient starvation or lack of growth factors that clears unnecessary intracellular organelles and proteins to the lysosome for degradation [41]. Therefore, autophagy is also important in protein homeostasis, sequestering and degrading long-life proteins, and invading microbes [41,[51][52][53]. Beside the classical degradative autophagy, a non-canonical function called secretory autophagy, which regulates unconventional secretory processes, is raising attention [54,55].…”
