Abstract. The senescence accelerated mouse prone (SAMP) mice with a shortened life span show accelerated changes in many of the signs of aging and have a shorter reproductive life span than SAM resistant (SAMR) controls. The reproductive senescence of SAMP is more accelerated than that of SAMR. In the present study, we found abnormal accumulation of luteal bodies (LBs) only in the ovaries of SAMP mice and, we examined the mechanism of abnormalities in luteal cell regression in the ovaries of SAMP mice. No significant changes were detected in serum progesterone or 17β-estradiol levels during estrus cycle between SAMP and SAMR mice. In abnormally accumulated LBs of SAMP mice, extremely high levels of activity of 20α-hydroxysteroid dehydrogenase, which catalyzes conversion of progesterone to the inactive form, 20α-hydroxyprogesterone were demonstrated histochemically. However, no marked differences in activity of 17β-hydroxysteroid dehydrogenase were detected histochemically. Moreover, no apoptotic cells were detected in the abnormally accumulated LBs of SAMP mice. Extremely weak immunohistochemical reactivity for endothelial nitric oxide synthase (eNOS) was noted in the abnormally accumulated LBs of SAMP mice, but no differences were seen in inducible NOS (iNOS). No marked differences in expression of eNOS-or iNOS-mRNA were seen during the estrus cycle in the ovaries of SAMP mice. However, high levels of expression of eNOS-mRNA at estrus stage and of iNOS-mRNA at estrus and metestrus stages were noted in the ovaries of SAMR mice. SAMP mice may be a useful model in which to examine the mechanism of regression of LBs in mammalian ovaries.

The present study was designed to investigate nitric oxide (NO) synthesis and the expression of endothelial NO synthase (eNOS) gene in cultured porcine granulosa cells. Granulosa cells prepared from small follicles (1-4 mm diameter) were cultured in plastic dishes coated with fibronectin in chemically defined medium, and matured after 48 h of stimulation with FSH. The concentrations of nitrite and nitrate remained relatively constant until 42 h of stimulation, after which they increased significantly up to twofold at 48 h. NO synthesis was accompanied by an increase in cGMP. Gene expression for eNOS was studied by RT-PCR, and a PCR product of the expected size amplified. eNOS mRNA was expressed in the presence of FSH, but not in the absence of FSH. Although eNOS mRNA was not expressed in the initial period, it was expressed after 12 h of stimulation with FSH, and remained at a relatively constant level until 48 h. Expression of eNOS mRNA preceded expression of LH receptor mRNA, which showed a maximal level at 24 h of stimulation. These observations suggest that eNOS expression is not related to a rapid synthesis of NO in developing granulosa cells, and that the activation of NO synthesis is rigidly regulated in the initial period of development.

By examining in vitro the effects of prostaglandin E-2 (PGE-2) and prostaglandin F-2 (PGF-2 ) induced in the corpora lutea (CL) of pseudopregnant rabbits, we have demonstrated that these prostaglandins modulate luteal nitric oxide synthase (NOS) activity and progesterone production differently, depending on the age of the CL. On CL obtained on day 4 of pseudopregnancy (day-4), PGE-2 was found to depress NOS total activity to 13% of control and to significantly increase basal progesterone secretion by 61%, while PGF-2 had no effect. On day-9 CL, PGE-2 was ineffective, but PGF-2 caused a 2·5-fold increase of NOS activity and a marked decrease in progesterone production. Using specific inhibitors, we found that the regulatory actions of PGE-2 in vitro are mediated via the adenyl cyclase/protein kinase A (PKA) second messenger system, while the PGF-2 -induced luteolytic effects on day-9 CL depend upon activation of the phospholipase C/protein kinase C (PKC) system. The different responsiveness of day-4 and day-9 CL to PGE-2 and PGF-2 could depend on receptor availability for these two prostaglandins, even if other cellular mechanisms cannot be excluded. The present study supports a functional role for NOS in regulating the steroidogenic capacity of rabbit CL, and reveals a novel interaction between a stimulatory G-protein-coupled receptor and PKC/PKA-mediated signal transduction modulating NOS activity.
