Regulation of Gap Junction Intercellular Communication in Primary Canine Lens Epithelial Cells: Role of Protein Kinase C

Abstract: Treatment with TPA significantly reduces GJIC in canine LEC. These effects are mediated, in part, by activation of calcium-dependent PKC isoforms. Primary canine LEC are a useful model in the study of the molecular mechanisms involved in GJIC and cataractogenesis.

“…As in these other systems, treatment of epithelial cells with phorbol esters (such as TPA) leads to a decrease in intercellular communication (81,131,159) associated with increased Cx43 phosphorylation (detectable by 32 P incorporation) (131). Several mechanisms have been proposed for the TPAinduced decrease in intercellular communication.…”

“…20) We had previously reported that treatment of PMA (10 nM) augmented MMP-9 activity in the human lens epithelial cell line, SRA01/04. 17) Therefore, we examined the effect of KPMFs on the PMA-mediated activities of gelatinase through the analysis of conditioned media from KPMFs-treated cells by gelatin zymography. As shown in Fig.…”

“…In lens epithelial cells, Cx43 is phosphorylated by PKC [198]. In a rabbit lens epithelail cell line [N/N1003A] it has been reported that Cx43 phosphorylation by PKC-gamma is important to prevent the spread of apoptosis in lenses under oxidative stress [199] due to a disassembly of gap junction plaques [200,201].…”

“…In a rabbit lens epithelail cell line [N/N1003A] it has been reported that Cx43 phosphorylation by PKC-gamma is important to prevent the spread of apoptosis in lenses under oxidative stress [199] due to a disassembly of gap junction plaques [200,201]. A decrease of gap junction communication induced by TPA [a PKC activator] was also observed in bovine [202] and canine [203] lens epithelial cells, where the zonula occludens protein-1 [ZO-1] appears to be essential for gap junction disassembly induced by PKC-gamma [204]. This TPA induced-phosphorylation was associated with resistance to degradation by a proteosome dependent pathway [205], and stimulate the interactions PKC-gamma with Cx43 and Cav-1 [206].…”
