Regulation of flavin-containing mono-oxygenase (Fmo3) gene expression by steroids in mice and humans

Abstract: Flavin-containing mono-oxygenases (FMOs) are a family of microsomal chemical- and drug-metabolizing enzymes. FMO3 is a major FMO form in adult mouse and human liver. FMO3 mutations have been associated with the incidence and severity of trimethylaminuria (TMAU), a metabolic disorder characterized by the inability of the affected individual to metabolize the odorous trimethylamine to its non-odorous N-oxide. In addition to this primary genetic form, there are other forms of TMAU that support the hypothesis that… Show more

“…Comparing male mice with intact testes to male mice that have been gonadectomized demonstrates that both hepatic FMO3 expression and TMAO generation are repressed by testosterone, while parallel studies in female mice demonstrate a positive correlation between estrogen and FMO3 gene expression. Furthermore, an estrogen receptor α binding site has been identified in the promoter region of FMO3 in both mouse and human samples [35]. Of note, the incidence of trimethylaminuria reported is higher among females, with patients reporting a worsening of symptoms occurring during menstruation [36].…”

“…TMA metabolism is regulated by a combination of genetic, dietary and hormonal factors, the complexity of which may obscure the interpretation of the precise roles of this pathway 14,15. For example, in humans, more than 30 FMO3 genetic variants have been reported to impair N -oxygenation of TMA and contribute to the fish odor syndrome phenotype 16.…”

“…Koukouritaki and others () determined that different haplotypes upstream of FMO3 significantly alter the promoter activity. In addition, FMO3 expression is regulated by steroids in humans and mice (Esposito and others ).…”
