Regulation of collagenase gene expression by IL–1β requires transcriptional and post-transcriptional mechanisms

Abstract: Interleukin-1 beta is believed to contribute to the pathophysiology of rheumatoid arthritis by activating collagenase gene expression. We have used a cell culture model of rabbit synovial fibroblasts to examine the molecular mechanisms of IL-1 beta-mediated collagenase gene expression. Stimulation of rabbit synovial fibroblasts with 10 ng/ml recombinant human IL-1 beta resulted in a 20-fold increase in collagenase mRNA by 12 h. Transient transfection studies using collagenase promoter-CAT constructs demonstrat… Show more

“…In most tumor cells, proMMP-1 mRNA levels are very low but can be induced by a wide variety of stimuli (7,8,(26)(27)(28). For instance, interleukin-1β, UV radiations or phorbol esters, increase proMMP-1 gene expression through histone posttranslational modifications (22,29).…”

“…Furthermore, MMP-1 overexpression is associated with several pathological conditions such as irreversible degradation of cartilage, tendon and bone in arthritis, and tumor invasion and metastasis (5-7). Transcriptional activation of proMMP-1 has been extensively studied, whereas the mechanisms involved in its expression remain largely unclear (7,8).…”

“…Furthermore, an earlier report demonstrated ERK1/2 activation alone potently up-regulates MMP-1 mRNA levels whereas p38 activation has no effect on the MMP-1 promoter (68). IL-1 itself has been shown to stabilize human collagenase mRNA (69). It is likely that IL-1 modulates MMP-13 production in an autocrine manner that augments the effects of serotonin and thus, amplifies the signaling cascade in myometrial SMCs.…”

“…GST subunits from untreated (C) or IL-1-treated (IL-1) (100 units/ml) hepatocytes were purified by glutathione-affinity column and separated by HPLC as described under "Experimental Procedures." the regulation of collagenase mRNA by IL-1 (35). However, in this case, this cytokine increased the stability of this transcript by reversing the destabilizing effects of sequences located in the 3Ј-untranslated regions.…”
