Regulation of cell death and survival in intestinal intraepithelial lymphocytes

Abstract: Intraepithelial lymphocytes (IEL) of the small murine bowel represent a unique population of mostly CD8 + T lymphocytes that reside within the epithelial cell layer of the intestinal mucosa. The close interaction with epithelial cells appears to be crucial for IEL survival since isolation and ex vivo culture induces massive apoptosis in this lymphocyte population. Here, we provide evidence that this form of IEL cell death may be mediated at least in part by endogenously produced glucocorticoids since adrenalec… Show more

“…In vitro exposure to GCs induces thymocyte apoptosis (18,26), and blocking GC receptors, inhibiting GC biosynthesis, and ADX reduce lymphocyte cell death (6,15,43). Furthermore, surgical removal of the adrenal glands is associated with less DNA fragmentation and lower nitric oxide production of splenic macrophages (5).…”

“…Tissue samples from duodenum and mid-jejunum were pooled for IEL isolation. The IEL were isolated by previously published protocols, which were adapted to canine lymphocytes [2,15]. In brief, IEL were dissociated from intestinal tissue by gently stirring tissue pieces in Ca 2+ -and Mg 2+ -free HBSS containing 2% horse serum, 1 mM DTT, and 0.5 mM EDTA for 2 · 30 min at 37°C.…”

“…In some experiments, splenocytes were pre-incubated with the GC receptor antagonist 100 nM RU486 (Brunner et al, 2001). After 2 h, cells were stimulated with 1 mg/ml concanavalin A for 20 h, and CD69 expression on CD4 þ T cells (antibodies from eBiosciences, San Diego, CA, USA) was detected by flow cytometry.…”
