Regulation of cell-cell interactions by phosphatidic acid phosphatase 2b/VCIP

Abstract: J.O.Humtsoe and S.Feng contributed equally to this workWe identi®ed vascular endothelial growth factor and type I collagen inducible protein (VCIP), also known as phosphatidic acid phosphatase 2b (PAP2b), in a functional assay of angiogenesis. VCIP/PAP2b exhibits an Arg±Gly±Asp (RGD) cell adhesion sequence. Immunoprecipitation and¯uorescence-activated cell sorting analyses demonstrated that VCIP-RGD is exposed to the outside of the cell surface. Retroviral transduction of VCIP induced cell aggregation/cell± ce… Show more

“…2E). LPP3 has recently been noted to contain an RGD sequence that interacts with ␤ 3 integrins and was therefore postulated to function in cell-cell interactions (26). Our results are consistent with an interaction between LPP3 and ␣ IIb ␤ 3 that may determine the lateral organization of LPP3 in the platelet membrane.…”

“…Effects of LPP3 inactivation in mice are similar to those seen when wnt signaling is up-regulated and LPP3 inhibits wnt signaling in cultured cells (27). LPP3 has also been shown to serve as an integrin ligand that regulates cell-cell interactions through a unique RGD sequence in the second extracellular loop (26). Clearly, more subtle approaches are required to separate activity-dependent and independent functions of these proteins.…”

“…VCIP is also known as a pro-angiogenic protein as addition of anti-VCIP antibody significantly blocked capillary morphogenesis of endothelial cells. 34,49,50 Because the efficiency of IRES-mediated translation is highly dependent on the cellular availability of IRES trans-acting factors, 44,51 and considering the important biological roles of VCIP, it is likely that the expression levels of VCIP's IRES trans-acting factors are consistently high in a broad range of cell types. Interestingly, VCIP IRES activity increases by 20-fold under hypoxic stress compared with normal cell culture conditions.…”

“…LPPs localized in the plasma membrane serve to attenuate the receptor-mediated signaling of extracellular LPA and S1P, which in turn activate a wide variety of signaling pathways such as PLC/ Ca ++ , Ras/ERK/MAPK, Rac, Rho and PI3K (Brindley, 2004, Moolenaar et al, 2004, Pyne et al, 2004. Furthermore, biological actions independent of their catalytic activity have been proposed for some LPPs (Escalante-Alcalde et al, 2003, Humtsoe et al, 2003, Zhao et al, 2005.…”
