Regulation of Carbohydrate Metabolism During Giardia Encystment1

Abstract: Giardia intestinalis trophozoites encyst when they are exposed to bile. During encystment, events related to the inducible synthesis of a novel N-acetyl-D-galactosamine (GalNAc) homopolymer, occur. Within the first 6 h of encystment, mRNA for glucosamine 6-P isomerase (GPI), the first inducible enzyme unique to this pathway appears, oxygen uptake rates double from non-encysting levels, and metronidazole (MTZ) inhibits oxygen uptake. Within 12 h, GPI and its activity are detectable and OU decreases 50% from non… Show more

“…The cyst wall is at least 40% protein, and the remainder is carbohydrate (14,15). The two known cyst wall proteins (CWPs) 1 contain similar domains, including a putative N-terminal signal peptide and five tandem, leucine-rich repeats (LRR) (16,17), and are reported to be phosphorylated (18).…”

“…All of these enzymes have been localized to the cytosol of encysting Giardia (Macechko et al, 1992). UDP-GalNAc is then converted into a unique b(1-3)-D-GalNAc homopolysaccharide by the action of a particle-associated b(1-3)GalNAc transferase, tentatively termed cyst wall synthase (CWS) (Jarroll et al, 2001). When trophozoites are induced to encyst, the first five of these enzymes are transcriptionally activated (Lopez et al, 2003) and the specific activities of all these enzymes increase (Macechko et al, 1992).…”
