Regulation of cancer‐immunity cycle and tumor microenvironment by nanobiomaterials to enhance tumor immunotherapy

Abstract: In the past decade, we have witnessed the revolution in cancer therapy, especially in the rapid development of cancer immunotherapy. In particular, the introduction of nanomedicine has achieved great improvement in breaking the limitations of and immunological tolerance caused by clinic‐approved immunotherapies (cancer vaccine, CAR‐T, and immune checkpoint blockade) to enhance immunogenicity, antigen presentation and T lymphocyte infiltration for eradicating the primary tumors and distant metastases simultaneo… Show more

“…In recent times, several cancer immunotherapies have been aimed at targeting the stages of the cancer immunity cycle. The latest advances in this research area include the application of nanomaterials for formulating improved cancer immunotherapies to achieve enhanced tumor-targeting and neutralizing the immuno-suppressive TME (Yang and Zhang, 2020). A recent work involving the fabrication of a three-in-one nanoplatform was successful in amplifying the various phases of the cancer-immunity cycle to boost the overall immune responses against cancer antigens.…”

“…Since the end of the twentieth century, nanomaterials have cut a striking figure in the biomedical field due to their attractive mechanical, optical, and electromagnetic properties in nanoscale that differ from traditional bulk materials (24). At present, nanomaterials together with mature modification technology have been widely used in medical imaging and disease diagnosis or treatment (25,26).…”

“…However, some patients did not respond to immunotherapy. The complexity and diversity of the tumor immune microenvironment (TIME) may determine its important impact on immunotherapy (Yang and Zhang 2020 ; Shang et al 2018 ). Immune cells and stromal cells are two important components of the extracellular matrix in the microenvironment.…”
