Regulated expression of the prolactin gene in rat pituitary tumor cells.

Abstract: Prolactin (PRL) gene expression in three strains of GH cells (rat pituitary tumor cells) has been quantitated by measurement of : (a) intracellular and extracellular PRL, (b) cytoplasmic translatable PRL-specific mRNA (mRNAP R L), and (c) molecular hybridization of cytoplasmic poly(A) RNA to cDNA PRL (DNA complementary to mRNA PRL) . Three GH cell lines utilized in this investigation were a PRL-producing (PRL + ) strain, GH 4C,, a PRL nonproducing 5-bromodeoxyuridine resistant (PRL -BrdUrd') strain, F,BGH,2C,,… Show more

“…The PRL production in GH4C1, 928-9b, and FBGH12C, before (GH4C1 > 928-9b > FBGH12C) and after TRH treatment could be correlated to the levels ofnuclear mRNAPRL sequences in these cells. A similar relationship was observed when cytoplasmic mRNAPRL sequences were examined in an analogous study (1). The increase in the amounts of nuclear RNAPRL sequences in TRH-treated cells as determined by RNA cDNAPRL solution hybridization would have been difficult to interpret without the blot analysis because of the possible contamination of nuclei with cytoplasm.…”

“…Previous studies have demonstrated a direct relationship between the amount of translatable mRNAPRL in control and TRH-or BrdUrd-treated cells and the levels of PRL production in each cell line (1,(4)(5)(6)(7). This direct relationship was also observed for the levels of hybridizable mRNAPRL sequences of control and TRH-treated cells (1,5). However, such studies did not show whether the variation in cytoplasmic mRNAPRL levels was a reflection ofnuclear events or ofalterations in cytoplasmic mRNAPRL turnover or stability.…”
