Regucalcin enhances adipocyte differentiation and attenuates inflammation in 3T3‐L1 cells

Abstract: Dysregulation of adipocyte differentiation and dysfunction play key roles in the pathogenesis of obesity and associated disorders such as diabetes and metabolic syndrome, and as such, a better understanding of the molecular mechanism of adipogenesis may help to elucidate the pathological condition of obesity and its associated disorders. Regucalcin (RGN) plays multiple regulatory roles in intracellular Ca 2+ signaling pathways in mammalian cells. Here, we report that overexpression of RG… Show more

“…In agreement with the above studies, our results showed that NM offers kidney protection through reducing the pro-inflammatory cytokines IL-6 and TNF-α in RIAKI. Our data showed that NM also upregulated the anti-inflammatory gene Rgn , which is consistent with the report that Rgn may act as a suppressor of inflammation in macrophage-infiltrated adipocyte tissue ( Murata et al, 2020 ). Surprisingly, we found that NM induced the expression of the pro-inflammatory chemokine Cxcl13.…”

“…The research evidence of RGN in immune microenvironment is relatively insufficient. A study has demonstrated that RGN could exert anti-inflammatory effects on adipocyte cocultured with macrophages 48 . Our results also observed that the high RGN expression group often showed the low level of macrophage infiltrations, including macrophage M0 and M1.…”

“…[17] Conventionally, impairment in adipocyte differentiation leads to obesity and insulin resistance. [18] 3T3-L1 preadipocytes differentiation is the well-established in vitro model for studying adipogenesis and an optimum platform to screen a variety of pro-adipogenic molecules that enhances differentiation at the cellular level. These cells were stimulated to differentiate in the presence of MDI (IBMX, DEXA, and Insulin cocktail) which is responsible for intracellular lipid accumulation.…”
