Registration of four upland cotton germplasm lines with elevated levels of seed oil oleic acid

Abstract: Four upland cotton (Gossypium hirsutum L.) germplasm lines, HOa1 (Reg. no. GP‐1072, PI 691652), HOa2 (Reg. no. GP‐1073, PI 691653), HOa3 (Reg. no. GP‐1074, PI 691654), and HOa4 (Reg. no. GP‐1075, PI 691655), with elevated levels of seed oil oleic acid were developed and released by the USDA‐ARS. High seed oil levels of oleic acid have been reported for a small number of G. barbadense L. accessions. Included among these accessions is GB713, which is also known as a source of reniform nematode (Rotylenchulus ren… Show more

“…todes, three lines exhibited some level of elevated oleic acid (Dowd et al, 2020). The M Rk-Rn 1 (PI 678938) and M Rk-Rn 5 (PI 678942) lines each exhibited elevated oleate levels in two of the five seeds tested with levels that varied between 31% and 44%.…”

“…One high-oleate accession of interest, GB713 (PI 608139), had also been reported to have high levels of resistance to reniform nematodes (Robinson et al, 2004) and had been used to breed this trait into the G. hirsutum cultivar Sure-Grow 747 (SG747, PVP 9800118) to produce two sets of nematode-resistant G. hirsutum plant lines (McCarty et al, 2013(McCarty et al, , 2017. Evaluation of individual seeds from these lines revealed elevated oleate levels in several seeds (Dowd et al, 2020). One of the three released M713Ren lines (M713Ren1, PI 665928) had elevated oleic acid levels (30%-35%) in two of the five seeds tested.…”

“…In addition, RNAi-directed down-regulation of PEPC2 up-regulated most lipid synthesis-related genes, resulting in 7.3% increase in cottonseed oil content ( Zhao et al, 2018a ). Shockey et al (2017) and Sturtevant et al (2017) identified a natural mutant allele of FAD2-1D in G. barbadense with high oleic acid in cottonseed oil, and its incorporation into G. hirsutum doubled oleic acid content ( Dowd et al, 2020 ). Most recently, Chen et al (2021) confirmed that knockout mutants of the GhFAD2 genes in Upland cotton by CRISPR/Cas9 editing increased the oleic acid level to 77.7% with a concomitant decrease in linoleic acid (from 58.6 to 6.9%) and palmitic acid (from 23.95 to 13.18%).…”

“…Four germplasm lines with elevated levels of seed oil oleic acid were developed and released in 2019 (Dowd et al, 2020). The lines were developed from previously released reniform nematoderesistant germplasm lines that were derived from G. barbadense accession GB713.…”
