Regional Heterogeneity in Murine Lung Fibroblasts from Normal Mice or Mice Exposed Once to Cigarette Smoke

Abstract: Chronic obstructive lung disease (COPD) is characterized by matrix deposition in the small airways but matrix loss from the parenchyma, phenomena which must depend on the ability of local fibroblasts to produce matrix after smoke exposure. To investigate this idea, we exposed C57Bl/6 mice once to cigarette smoke or to air (control) and prepared primary cultures of lung fibroblasts by microdissecting large airways (trachea, LAF), medium size airways (major bronchi, MAF) and parenchyma (PF). Control PF showed th… Show more

“…For direct instillation, cells were detached with trypsin, washed, and re-suspended in PBS. For in vitro internalization essays, lung fibroblasts were obtained from healthy C57BL/6 mice and cultivated in DMEM containing 1% antibiotic solution, 10% fetal bovine serum (FBS) and 15 mM HEPES for up to 2 weeks [ 27 ]. MH-S cells were purchased from ATCC (#CRL2019) and cultivated in RPMI 1640 with 10% FBS, 1% penicillin-streptomycin (10,000 U/mL, Thermo Fisher Scientific, USA) and 5 mM 2-mercaptoethanol according to the vendor’s instructions.…”

“…DNA methylation is an epigenetic modification of DNA that regulates gene expression through transcriptional regulation. It is mediated by DNA methyltransferases (DNMTs) which catalyze the transfer of methyl groups to the cytosine residue in DNA . There are 4 known DNMTs at present, DNMT1, 2, 3a, and 3b.…”

Among all DNMTs, the main function of DNMT1 is to maintain DNA methylation (also known as maintenance of methyltransferase).…”

“…While there was more biglycan production from distal lung fibroblasts than central fibroblasts derived from normal subjects, this heterogeneity was not seen in fibroblasts from COPD subjects . Finally, Preobrazhenska et al reported regional heterogeneity in murine lung fibroblasts with regard to matrix production and wound repair under basal conditions or after in vivo cigarette smoke exposure .…”
