Regional and subtype selective changes of neurotransmitter receptor density in a rat transgenic for the Huntington's disease mutation

Abstract: Huntington's disease (HD) is an autosomal dominantly inherited progressive neurodegenerative disorder caused by a CAG/polyglutamine repeat expansion in the gene encoding the huntingtin protein. We have recently generated a rat model transgenic for HD, which displays a slowly progressive phenotype resembling the human adult-onset type of disease. In this study we systematically assessed the distribution and density of 17 transmitter receptors in the brains of 2-year-old rats using quantitative multi-tracer auto… Show more

“…Furthermore, a reduction in striatal D1-receptor density was found in 14 months old tgHD rats (Bode et al, 2008), as well as a decrease in striatal D1 and D2 receptor binding at the age of 24 months (Bauer et al, 2005; Von Hörsten et al, 2003). A reduction of serotonin-containing cells and an increase of dopamine-containing cells were also found in the dorsal raphe nucleus in both human and tgHD rat tissues, inducing a reduced level of serotonin expression in the medial PFC (Jahanshahi et al, 2013).…”

“…Furthermore, the desensitization of the 5-HT 1A autoreceptor has been demonstrated to be essential for enhanced 5-HT transmission following chronic treatment with selective 5-HT reuptake inhibitors [21]. Finally, 5-HT 1A and/or 5-HT 2 receptor abnormalities have been described in different rodent models of mood disorders [22], [23], [24], [25] and HD [26], [27], [28].…”

“…Tissue sections were dried under a gentle stream of cool air. Slides were arrayed in X-ray cassettes together with tritium standards (Amersham Pharmacia Biotech, Piscataway, NJ, USA) and were exposed to tritium-sensitive film (Kodak hyperfilm; Eastman Kodak) at room temperature for 2 or 3 months (1315). Films were developed and fixed at room temperature (Table I).…”
