In vitro growth of Quercus robur L. buds taken from 1-7 month old seedlings or adult plant material was studied. The following were investigated for their effects on the establishment of explants and subsequent subcul~res: original position and lignification of the primary explant, conditioning and ageing of source plants, incorporation of N 6 benzyladenine (BA) and activated charcoal in the medium.For bud break the best results were obtained with explants from herbaceous twigs in all tested media. For shoot growth the results depended on the medium used. Medium containing activated charcoal produced episodic growth, leaf organogenesis was reduced, spontaneous rooting occurred, but subculturing from adult plant material failed. On medium containing more than 8.8/aM of BA, all the buds developed abnormally and elongation did not occur. At lower concentrations of BA (4.4/aM) shoots elongated, leaf organogenesis was increased and episodic growth tended to disappear on subcultured seedling explants. No spontaneous rooting was observed, but subculture from adult plant material was managed successfully.

Multiplication végétative du Weigela florida "Bristol Huby" à partir d'apex inflorescentiels en culture in vitro (*) par Eleni MALOUPA-IKONOMOU e) L'induction florale des plantes-mères est réalisée par un arrêt de croissance provoqué par un passage de 3 semaines en jours courts dont la nuit longue est interrompue par 3 fois 30 minutes de rouge sombre. Pour obtenir le maximum d'organes végétatifs,les apex inflorescentiels mis en culture in vitro doivent âtre prélevés à un stade précis de leur développement tel que seuls les sépales soient visibles sur les boutons les plus avancés. Summary.-ln vitro vegetative multiplication of Weigela was carried out via the culture of floral apices excised at a specifie floral initiation stage.Floral induction was produced by an arrest of growth induced by a stay of 3 weeks in short days with night interrupted by 3 far red night breaks was the best floral induction pattern. The maximal floral d~ferenciation with a good muhiplication rate was etablished with sapai initiation stage for the most differenciated buds.We obtained at !east tan plants from a single floral meristem cuhured on A. Duran medium {1975) supplemented with vitamins and active charcoal. lt is interesting ta note that the meristem explants remain vegetative at 16 h photoperiod whereas nodal stem cuttings become floral.

The effect of long-term in vitro shoot culture on somatic embryogenesis in quince BA 29 was investigated. Three experiments were performed on leaves explanted at about 8-mo. intervals from the same culture stock and maintained under different light qualities. Embryo production was assessed either in terms of percentage of embryogenic leaves or number of embryos per leaf. By appropriate data processing both these responses were linearly related to photoequilibrium in each experiment. Statistical comparisons among the three experiments showed significant differences both in mean (computed over light qualities) and line slope values. In particular, with increasing shoot culture age, both percentage of embryogenic leaves and number of embryos per leaf progressively increased, while mean slope values decreased. The increase in mean values suggests a positive effect on somatic embryogenesis due to possible tissue rejuvenation when mother cultures were cultivated in vitro for longer periods. Slope decrease over time indicated the interactions between age of the in vitro culture and photoequilibrium. Thus, embryo production at different culture ages was consistently found to be highest at high photoequilibrium values; in contrast, if a low level of phytochrome was activated, embryogenesis in the youngest cultures was low or absent, but increased with the progressive tissue rejuvenation arising from long-term in vitro culture
