Refined approach for quantification of in vivo ischemia-reperfusion injury in the mouse heart

Abstract: Cardiac ischemia-reperfusion experiments in the mouse are important in vivo models of human disease. Infarct size is a particularly important scientific readout as virtually all cardiocirculatory pathways are affected by it. Therefore, such measurements must be exact and valid. The histological analysis, however, remains technically challenging, and the resulting quality is often unsatisfactory. For this report we have scrutinized each step involved in standard double-staining histology. We have tested publish… Show more

“…Myocardial infarct size was determined by using Evans blue/2,3,5-triphenyltetrazolium chloride (TTC) staining as described by Bohl et al (27). Briefly, the ascending aorta was cannulated with a 20-gauge tubing adapter, and 1% Evans blue was perfused into the aorta and coronary arteries to delineate the "area at risk" (AAR).…”

“…Myocardial I/R mouse model and infarct size determination were performed as previously reported (48). PA28α/tTA double-Tg mice and littermate tTA single-Tg mice at 8 weeks old were anesthetized with 1% isoflurane and ventilated through intubation via mouth.…”
