Reference materials for MS-based untargeted metabolomics and lipidomics: a review by the metabolomics quality assurance and quality control consortium (mQACC)

Abstract: Introduction The metabolomics quality assurance and quality control consortium (mQACC) is enabling the identification, development, prioritization, and promotion of suitable reference materials (RMs) to be used in quality assurance (QA) and quality control (QC) for untargeted metabolomics research. Objectives This review aims to highlight current RMs, and methodologies used within untargeted metabolomics and lipidomics communities to ensure standar… Show more

“…Furthermore, targeted investigations only look for a limited number of metabolites but with the respective standard substances. As opposed to this, untargeted approaches search for differences in an undefined set of molecules without the possibility of applying internal standards for every compound [ 8 ]. Thus, the search for biomarkers for a certain pathological disorder is highly dependent on the methodology used.…”

“…Samples were randomly analyzed by UPLC-QTOF-MS within a template of intrastudy quality control (QC) samples in both analytical methods. Since the lipid profiling study was conducted before the metabolic profiling study, additional QC practices were implemented for the latter to narrow down the number of analytically robust features for data analysis and interpretation . For lipid profiling, 10 intrastudy QC samples consisting of pools of equal volume aliquots of subsets of the study samples were generated and followed protein precipitation and further sample preparation steps.…”

“…However, the fundamental efforts to establish analytical consensus values in reference materials (e.g., NIST SRM lipidomics community (http://lipidomics-standards-initiative.org) 3,16,17,18,19,20 . The systematic analysis of shared reference materials has emerged as the best normalization strategy to correct method-specific biases (e.g., application of different extraction procedures and analytical platforms for lipid measurement) 21,22,23,24,25 . Thus, the time is right to foresee the collection of biologically relevant data necessary to establish the reference intervals for populations with different ethnic backgrounds and dietary regimens across different geographical regions.…”
